Legacy and emerging per- and polyfluoroalkyl substances (PFAS) regulate steroidogenesis in the male gonad
Endocrinology. 2023 Sep 28:bqad142. doi: 10.1210/endocr/bqad142. Online ahead of print.ABSTRACTPer- and polyfluoroalkyl substances (PFAS) are widely used in a variety of industrial processes and manufacturing of consumer products. Current efforts by the manufacturing industry will limit use of long-chain or legacy PFAS represented by perfluorooctanoic acid (PFOA) and perfluorooctane sulfonic acid (PFOS) and replace with short-chain or emerging PFAS such as perfluorobutanoic acid (PFBA) and perfluorobutane sulfonic acid (PFBS). However, there is little to no information on the toxicity of new and emerging PFAS. Therefore, w...

Functional Water Channels Within the TSH Receptor - a new paradigm for TSH action with disease implications
Endocrinology. 2023 Sep 28:bqad146. doi: 10.1210/endocr/bqad146. Online ahead of print.ABSTRACTThe TSH receptor (TSHR) transmembrane domain (TMD) lives in the plasma membrane consisting of lipids and water molecules. To understand the role of TSHR-associated water molecules we used molecular dynamic simulations of the TMD and identified a network of putative receptor associated transmembrane water channels. This result was confirmed with extended simulations of the full length TSHR with and without TSH ligand binding. While the transport time observed in the simulations via the TSHR protein was slower than via the lipid bi...

Puberty suppression followed by testosterone therapy does not impair reproductive potential in female mice
This study used a translational murine IVF model to examine the effects of female puberty suppression with depot leuprolide acetate (LA), followed by virilizing therapy with testosterone cypionate (T), on embryologic and pregnancy outcomes. LA effectively inhibited puberty when mice were treated beginning at 3 weeks of age. LA treatment was associated with higher mouse weight but lower ovarian weight. LA-treated mice ovulated developmentally competent eggs in response to gonadotropin administration, albeit at a higher dose than controls. Ovaries from mice treated with LA and T produced oocytes that had morphologically norm...

Research Progress of Metformin in the Treatment of Oral Squamous Cell Carcinoma
Endocrinology. 2023 Sep 20:bqad139. doi: 10.1210/endocr/bqad139. Online ahead of print.ABSTRACTOral squamous cell carcinoma (OSCC) is one of the most common malignancies with high mortality, posing a great threat to both human physical and mental health. With the advancement of scientific research, a variety of cancer therapies have been used for OSCC treatment. However, the prognosis of OSCC shows no significant improvement. Metformin has been recognized as the first-line drug for the treatment of diabetes, and recent studies have shown that metformin has a remarkable suppressive effect on tumor progression. Metformin can...

Adipocyte glucocorticoid receptor inhibits immune regulatory genes to maintain immune cell homeostasis in adipose tissue
Endocrinology. 2023 Sep 20:bqad143. doi: 10.1210/endocr/bqad143. Online ahead of print.ABSTRACTGlucocorticoids acting via the glucocorticoid receptors (GR) are key regulators of metabolism and the stress response. However, uncontrolled or excessive GR signaling adversely affects adipose tissue, including endocrine, immune, and metabolic functions. Inflammation of the adipose tissue promotes systemic metabolic dysfunction; however, the molecular mechanisms underlying the role of adipocyte GR in regulating genes associated with adipose tissue inflammation are poorly understood. We performed in vivo studies using adipocyte sp...

Neuronal AR regulates glucose homeostasis and energy expenditure in lean female mice with androgen excess
Endocrinology. 2023 Sep 21:bqad141. doi: 10.1210/endocr/bqad141. Online ahead of print.ABSTRACTHyperandrogenemia and polycystic ovarian syndrome are a result of the imbalance of androgen levels in females. Androgen receptor (Ar) mediates the effect of androgen, and this study examines how neuronal Ar in the central nervous system (CNS) mediates metabolism under normal and increased androgen conditions in female mice. The neuron specific ARKO mouse (SynARKO) was created from female (Ar fl/wt; synapsin promoter driven Cre) and male (Ar fl/y) mice. Glucose tolerance test revealed impaired glucose tolerance that was partially ...
