DHT and insulin up-regulate secretion of the soluble decoy receptor of IL-33 from decidualized endometrial stromal cells
Endocrinology. 2023 Nov 16:bqad174. doi: 10.1210/endocr/bqad174. Online ahead of print.ABSTRACTInterleukin 33 (IL-33) signaling regulates most of the key processes of pregnancy, including decidualization, trophoblast proliferation and invasion, vascular remodeling, and placental growth. Accordingly, dysregulation of IL-33, its membrane bound receptor (ST2L, transducer of IL-33 signaling) and its soluble decoy receptor (sST2, inhibitor of IL-33 signaling) has been linked to a wide range of adverse pregnancy outcomes that are common in women with obesity and polycystic ovary syndrome, i.e., conditions associated with hyperan...

Fibroblast Growth Factor 23 Neutralizing Antibody Ameliorates Abnormal Renal Phosphate Handling in Sickle Cell Disease Mice
Endocrinology. 2023 Nov 2;164(12):bqad173. doi: 10.1210/endocr/bqad173.ABSTRACTWe assessed the involvement of fibroblast growth factor 23 (FGF23) in phosphaturia in sickle cell disease (SCD) mice. Control and SCD mice were treated with FGF23 neutralizing antibody (FGF23Ab) for 24 hours. Serum ferritin was significantly increased in SCD mice and was significantly reduced in female but not male SCD mice by FGF23Ab. FGF23Ab significantly reduced increased erythropoietin in SCD kidneys. Serum intact FGF23 was significantly increased in SCD female mice and was markedly increased in SCD male mice; however, FGF23Ab significantly ...

Increased OCT3 expression in adipose tissue with aging: implications for lipid turnover and insulin resistance in women
CONCLUSION: OCT3 mRNA and protein levels in SAT were increased with aging, and mRNA levels were negatively associated with markers of insulin resistance. E2 incubation downregulated OCT3 mRNA levels, which may explain lower OCT3 mRNA levels in premenopausal compared to postmenopausal women. High OCT3 mRNA and protein levels may result in increased catecholamine degradation, which may account for the reduction in adipose tissue lipolysis observed with aging.PMID:37972266 | DOI:10.1210/endocr/bqad172 (Source: Endocrinology)

Top-Down Proteomics of Mouse Islets With Beta Cell CPE Deletion Reveals Molecular Details in Prohormone Processing
Endocrinology. 2023 Nov 2;164(12):bqad160. doi: 10.1210/endocr/bqad160.ABSTRACTAltered prohormone processing, such as with proinsulin and pro-islet amyloid polypeptide (proIAPP), has been reported as an important feature of prediabetes and diabetes. Proinsulin processing includes removal of several C-terminal basic amino acids and is performed principally by the exopeptidase carboxypeptidase E (CPE), and mutations in CPE or other prohormone convertase enzymes (PC1/3 and PC2) result in hyperproinsulinemia. A comprehensive characterization of the forms and quantities of improperly processed insulin and other hormone products...

Insights into RNA N6-methyladenosine in glucose and lipid metabolic diseases and their therapeutic strategies
Endocrinology. 2023 Nov 9:bqad170. doi: 10.1210/endocr/bqad170. Online ahead of print.ABSTRACTThe incidence of glucose and lipid metabolism diseases, including type 2 diabetes, obesity, metabolic syndrome, and nonalcoholic fatty liver disease is rising, which places an enormous burden on people around the world. However, the mechanism behind these disorders remains incompletely understood. N6-methyladenosine (m6A) is one type of posttranscriptional RNA modification and research has shown that it plays a crucial role in several metabolic diseases. m6A methylation is reversibly and dynamically regulated by methyltransferases...

Progesterone activates histone lactylation-Hif1 α-glycolysis feedback loop to promote decidualization
Endocrinology. 2023 Nov 10:bqad169. doi: 10.1210/endocr/bqad169. Online ahead of print.ABSTRACTDecidualization is a progesterone-dependent cellular differentiation process that is essential for establishing pregnancy. Robust activation of glycolysis and lactate synthesis during decidualization is remarkable, but their developmental functions remain largely unknown. Herein, we identify that endometrial lactate production plays a critical role in establishing local histone lactylation, a newly identified histone modification, and is important for ensuring normal decidualization. Enhanced endometrial glycolysis is the hallmar...
