Screening system for EGFR mutation detection in cytology cell ‐free DNA of cerebrospinal fluid based on assured sample quality
AbstractBackgroundThe cobas® EGFR Mutation Test v2 designed for cell ‐free DNA (cfDNA) is approved as a companion diagnostic for osimertinib therapy. The aim of this study was to evaluate the concordance ofEGFR mutation detection between paired primary or recurrent samples, and cerebrospinal fluid (CSF) cytology samples of lung cancer patients.MethodsA total of 26 lung cancer patients with supernatant cytology cfDNA in CSF was analyzed forEGFR mutations using the cobas® EGFR Mutation Test v2.0 designed for cfDNA, and the concordance rates between CSF cfDNA and primary or recurrent samples were investigated.Result...

Liquid ‐based endometrial cytology using SurePathTM is not inferior to suction endometrial tissue biopsy for detecting endometrial malignancies‐Midterm report of a multicenter study advocated by Japan Association of Obstetricians and Gynecologists‐
ConclusionsThe clinical performance of liquid ‐based endometrial cytology for detecting endometrial malignancies was almost identical to the performance of suction endometrial tissue biopsy. This indicates that liquid‐based endometrial cytology was not inferior to suction endometrial tissue biopsy for the detection of endometrial cancer. Th e liquid‐based endometrial cytology is appropriate for various clinical situations as the first‐step detecting tool. In addition, it could be used for cancer surveillance for women with signs highly suggestive of endometrial malignancies and in Lynch syndrome patients, on a larg...

Abnormal Immunolabeling of SMAD4 in Cell Block Specimens Distinguish Malignant and Benign Pancreatic Cells
ConclusionsDecreased SMAD4 immunolabeling provided improved sensitivity as compared to negative SMAD4 immunolabeling; therefore, it is important to compare SMAD4 expression in a sample to its expression in normal cells. Abnormal SMAD4 labeling showed low sensitivity and high specificity; therefore, SMAD4 staining using EUS ‐FNA samples might be helpful to detect malignancies that possessSMAD4 gene abnormalities. This article is protected by copyright. Diagnostic utility and pitfalls of intraoperative pulmonary imprint cytology based on final pathological diagnoses
ConclusionIC ‐based diagnoses of pulmonary lesions as malignant corresponded well with the final histopathological diagnoses, but IC‐based diagnoses of negative (i.e., without malignant cells) were not as reliable. Thus, pathologists should recognize the limitations of IC, especially for identifying malignan t lesions. Also, the possibility of latent bacterial infection in a granuloma with caseous necrosis indicates that an IC preparation deemed inadequate for diagnosis should not be ignored. This article is protected by copyright. Comparison of the Cervex ‐Brush alone to Cytobrush plus Cervex‐Brush for detection of cervical dysplasia in women with a transformation zone type 3
This study examines the effectiveness of a cytobrush in addition to a standard Cervex‐Brush when used in TZ3 assessment.MethodologyAn 18 month diagnostic accuracy study in a single NHS Trust. Women with a TZ3 booked for LLETZ with a referral cytology of high risk HPV and squamous dyskaryosis were recruited. Immediately prior to LLETZ a Cervex ‐Brush plus Cytobrush liquid‐based cytology sample was taken. Presence of endocervical cells were compared. Predictability of CIN2+ was by blind standardised reporting of the LLETZ histology.ResultsOne hundred and five women were recruited from a total eligible population of 153...

Noninvasive follicular thyroid neoplasm with papillary ‐like nuclear features: An interobserver study of key cytomorphological features from a large academic medical centre
AbstractObjectiveBecause of the indolent nature of noninvasive follicular thyroid neoplasm with papillary ‐like nuclear features (NIFTP) and potential requisite for a more conservative treatment, it is crucial to identify features of this entity preoperatively. Our group recently published findings that there are several cytomorphological features that may be used as clues to distinguish NIFTP, papill ary thyroid carcinoma (PTC) and follicular adenoma (FA) on fine needle aspiration. Therefore, we aimed to determine the interobserver reproducibility of these findings.MethodsPresurgical fine ‐needle aspiration slides fro...

Clinicocytopathological spectrum, including uncommon forms, of nine cases of chordomas with immunohistochemical results, including brachyury immunostaining: A single institutional experience
ConclusionsChordomas can be primarily diagnosed by fine needle aspiration cytology in a typical clinicoradiological setting with a combination ofkey cytomorphological features. Pleomorphic cells and intranuclear inclusions are associated with a relatively aggressive subtype. An exact diagnosis has treatment implications and requires confirmation by brachyury immunostaining.
