A Quinquennial Review on Recent Advancements and Developments in Search of Anti- malarial Agents
Curr Top Med Chem. 2023 Apr 27. doi: 10.2174/1568026623666230427115241. Online ahead of print.ABSTRACTMalaria has been a major parasitic disease in tropical and subtropical regions and is estimated to kill between one and two million people (mainly children) every year. Novel anti-malarial agents are urgently needed to combat the malarial parasites enduring resistance to the current medications, leading to increased morbidity and mortality. The heterocycles, holding a prominent position in chemistry and found in both natural and synthetic sources, have shown several biological activities including anti-malarial activity. T...

Review of the Application of PAT in the Pharmaceutical Continuous Crystallization Process
Curr Top Med Chem. 2023 Apr 20. doi: 10.2174/1568026623666230420112709. Online ahead of print.ABSTRACTAs an important pharmaceutical process, crystallization greatly impacts the final product. In recent years, the continuous crystallization process has attracted more attention from researchers, with the promotion of continuous manufacturing (CM) by the Food and Drug Administration (FDA). The continuous crystallization process has the advantages of high economic benefit, stable and uniform quality, a short production cycle, and personalization. To carry out continuous crystallization, some related process analytical technol...

Natural products as antifungal agents against invasive fungi
CONCLUSION: In this review, we attempted to review the available literature on natural antifungal products and their derivatives. Most of the studied compounds showed potent activity against Candida species, Aspergillus species, or Cryptococcus species. Some of the studied compounds also demonstrated the ability to impair the cell membrane and cell wall, inhibit hypha and biofilms, and cause mitochondrial dysfunction. Although the MoAs of these compounds are not well understood yet, they can be used as lead components for the development of new, effective, and safe antifungal agents through their novel mechanisms.PMID:3707...

Current Medicinal Insights on Synthetic Small Molecules and Natural Origin Products as PD-1/PD-L1 Inhibitors
Curr Top Med Chem. 2023 Apr 17. doi: 10.2174/1568026623666230417111616. Online ahead of print.ABSTRACTCancer is one of the leading causes of death worldwide. Each year, millions of people are diagnosed with cancer; hence, researchers have always been curious and busy developing cancer treatments. Despite thousands of studies, cancer is still a major threat to human beings. One of the mechanisms through which cancer invades a human being is the immune escape mechanism, which has been the focus of studies in the past years. PD-1/PD-L1 pathway plays a major role in this immune escape. Therefore, research focusing on blocking ...

Advances in Drug Discovery Based on Genomics, Proteomics and Bioinformatics in Malaria
Curr Top Med Chem. 2023 Apr 18. doi: 10.2174/1568026623666230418114455. Online ahead of print.ABSTRACTMalaria is one of the neglected infectious diseases, and drugs are the first line of action taken against the onset of malaria as therapeutics. The drugs can be of either natural or artificial origin. Drug development has multiple impediments grouped under three categories, a. drug discovery and screening, b. the drug's action on the host and the pathogen, and c. clinical trials. Drug development takes coon's age from discovery to the market after FDA approval. At the same time, targeted organisms develop drug resistance q...
