Pharmacogenetics of tamoxifen in breast cancer patients of African descent: Lack of data
Clin Transl Sci. 2024 Mar;17(3):e13761. doi: 10.1111/cts.13761.ABSTRACTTamoxifen, a selective estrogen receptor modulator, is used to treat hormone receptor-positive breast cancer. Tamoxifen acts as a prodrug, with its primary therapeutic effect mediated by its principal metabolite, endoxifen. However, tamoxifen has complex pharmacokinetics involving several drug-metabolizing enzymes and transporters influencing its disposition. Genes encoding enzymes involved in tamoxifen disposition exhibit genetic polymorphisms which vary widely across world populations. This review highlights the lack of data on tamoxifen pharmacogenet...

Optimizing smartphone psychotherapy for depressive symptoms in patients with cancer: Multiphase optimization strategy using a decentralized multicenter randomized clinical trial (J-SUPPORT 2001 Study)
CONCLUSION: As the presence of any of the three intervention components did not contribute to a significant additive reduction of depressive symptoms, we cannot make evidence-based recommendations regarding the use of specific smartphone psychotherapy.PMID:38468404 | DOI:10.1111/pcn.13657 (Source: Clinical Genitourinary Cancer)

Lifestyle Factors and Cancer: A Narrative Review
Mayo Clin Proc Innov Qual Outcomes. 2024 Mar 4;8(2):166-183. doi: 10.1016/j.mayocpiqo.2024.01.004. eCollection 2024 Apr.ABSTRACTLifestyle factors and their impact on cancer prevention, prognosis, and survivorship are increasingly recognized in the medical literature. Lifestyle factors are primarily defined here as diet and physical activity. We conducted a narrative review of the primary published data, including randomized controlled trials and prospective studies, on the impact of primary lifestyle factors on oncogenesis and clinical outcomes in the preventative and survivorship setting. First, we discuss the oncogenic m...

Cell-free DNA concentration as a biomarker of response and recurrence in HER2-negative breast cancer receiving neoadjuvant chemotherapy
CONCLUSIONS: In TNBC, cfDNA concentrations during therapy were not strongly correlated with response or prognosis. In the HR-positive/HER2-negative group, pretreatment cfDNA concentration was prognostic for DRFS. Overall, the predictive and prognostic value of cfDNA concentration was more limited than that of ctDNA.PMID:38470545 | DOI:10.1158/1078-0432.CCR-23-2928 (Source: Clinical Genitourinary Cancer)

A novel body composition risk score (B-Score) and overall survival among patients with nonmetastatic breast cancer
CONCLUSIONS: More unfavorable body composition characteristics were associated with increased risks of overall mortality in a dose-response manner. Considering body composition measurements together as a composite score (B-Score) may improve risk stratification and inform dietary and lifestyle interventions following breast cancer diagnosis.PMID:38471402 | DOI:10.1016/j.clnu.2024.03.001 (Source: Clinical Genitourinary Cancer)

The Clinicopathological and Prognostic Significance of HER2-Low Breast Cancer: A Comparative Analysis Between HER2-Low and HER2-Zero Subtypes
CONCLUSION: HER2-low accounted for 70% of the cases and correlated with favorable biological markers. The HER2-low group had a significantly better OS than the HER2-0 group. However, the response to NAC was low in the HER2-low group, and this group had the poorest prognosis among all the non-pCR cases. These findings indicate that HER2-low may have a different biology and prognosis and therefore should be classified as a new entity.PMID:38472058 | DOI:10.1016/j.clbc.2024.02.013 (Source: Clinical Genitourinary Cancer)

Tumor deposits in axillary adipose tissue in patients with breast cancer: Do they matter?
World J Clin Cases. 2024 Feb 26;12(6):1045-1049. doi: 10.12998/wjcc.v12.i6.1045.ABSTRACTTumor deposits (TDs) are defined as discrete, irregular clusters of tumor cells lying in the soft tissue adjacent to but separate from the primary tumor, and are usually found in the lymphatic drainage area of the primary tumor. By definition, no residual lymph node structure should be identified in these tumor masses. At present, TDs are mainly reported in colorectal cancer, with a few reports in gastric cancer. There are very few reports on breast cancer (BC). For TDs, current dominant theories suggest that these are the result of lym...

The influence of marital status at diagnosis on survival of adult patients with mantle cell lymphoma
CONCLUSION: Marital status at diagnosis is an independent prognostic factor for MCL patients, with widowed individuals showing worse OS and CSS than those who are married, single, or divorced/separated. Adequate psychological and social support for widowed patients is crucial for improving outcomes in this patient population.PMID:38466431 | PMC:PMC10927831 | DOI:10.1007/s00432-024-05647-z (Source: Clinical Genitourinary Cancer)

Molecular probes targeting HER2 PET/CT and their application in advanced breast cancer
CONCLUSION: HER2-targeted PET/CT offers a promising noninvasive approach for real-time assessment of HER2 status, which can be guide targeted treatment for HER2-positive breast cancer patients. Future prospective clinical studies will be invaluable for fully evaluating the importance of HER2-targeted molecular imaging in the management of breast cancer.PMID:38466436 | PMC:PMC10927773 | DOI:10.1007/s00432-023-05519-y (Source: Clinical Genitourinary Cancer)

Tumor-infiltrating lymphocytes refine outcomes in triple-negative breast cancer treated with anthracycline-free neoadjuvant chemotherapy
CONCLUSIONS: sTILs density was independent predictor of OS beyond clinicopathologic features and pathologic response in TNBC patients treated with anthracycline-free CbD chemotherapy. Notably, sTILs density stratified outcomes beyond TNM stage and pathologic response. These findings highlight role of sTILs in patient selection/stratification for neo/adjuvant escalation and de-escalation strategies.PMID:38466643 | DOI:10.1158/1078-0432.CCR-24-0106 (Source: Clinical Genitourinary Cancer)
