Do metabolites account for higher serum steroid hormone levels measured by RIA compared to mass spectrometry?
This study aimed to determine which estradiol metabolites may contribute to this effect. LC-MS/MS analysis was performed on 70 serum samples from premenopausal women, after purification by extraction and Celite column partition chromatography as would be used prior to conventional RIA for estradiol. The metabolites estrone and 2-methoxyestradiol accounted for 6.9% and 2.1% of the estradiol fractions in the purified samples overall, but the extent of contamination with these metabolites was greater in the samples containing 50 pg/mL estradiol. However, since these metabolites have a

Urinary N-terminal fragment of titin is a marker to diagnose muscular dystrophy in patients with cardiomyopathy
Publication date: September 2018Source: Clinica Chimica Acta, Volume 484Author(s): Akiomi Yoshihisa, Takatoyo Kiko, Takamasa Sato, Masayoshi Oikawa, Atsushi Kobayashi, Yasuchika TakeishiAbstractThe differential diagnosis of cardiomyopathy is important. It has been recently reported that urinary titin N (U-TN) is increased in patients with muscular dystrophy (MD), and is associated with muscular damage. We aimed to clarify whether U-TN is useful as a diagnostic tool for distinguishing MD from various cardiomyopathies [e.g. dilated cardiomyopathy (DCM), hypertrophic cardiomyopathy (HCM)]. We measured and compared the U-TN/cr...

Fake news? Biotin interference in thyroid immunoassays
We report on a 47 year old male patient with multiple sclerosis (MS) presenting in our outpatient neurology clinic in Frankfurt/Main for therapy evaluation. Before change of treatment laboratory investigations were performed. Thyroid function tests (TFTs) with a streptavidin/biotin based immunoassay revealed severe hyperthyroidism with positive thyroid autoantibodies suggestive for Graves' disease. Clinical presentation and thyroid sonography were unremarkable. Due to the discordance between clinical presentation and TFTs, we repeated medical history, in which the patient reported taking high-doses of biotin (300 mg/da...

Hypothesis: Postprandial remnant lipoproteins are the causal factors that induce the insulin resistance associated with obesity
Publication date: October 2018Source: Clinica Chimica Acta, Volume 485Author(s): Katsuyuki Nakajima, Yoshiharu Tokita, Akira TanakaAbstractWe have long thought that remnant lipoproteins (RLP) in plasma are significantly increased as the result of disturbed lipoprotein metabolism followed by obesity and insulin resistance. Therefore, it was believed that insulin resistance causes and enhances RLP formation. In contrast, this hypothesis states that RLP induces insulin resistance as the result of obesity associated with the excessive fat intake. The majority of plasma TG increased after fat intake is TG in RLP (RLP-TG) and th...

Assessing the stability of Cd, Mn, Pb, Se, and total Hg in whole human blood by ICP-DRC-MS as a function of temperature and time
ConclusionsStorage temperatures of −20 °C and 4 °C are equivalent to −70 °C for stability of Cd, Mn, Pb, Se, and Hg in human whole blood for at least 36 months when blood is stored in sealed polypropylene vials. Increasing the sample storage temperature from −70 °C to −20 °C or 4 °C can lead to large energy savings. The best analytical results are obtained when storage time at higher temperature conditions (e.g. 23 °C and 37 °C) is minimized because recovery of Se and Hg is reduced. Blood samples stored in polypropylene cryovials also lose volume ove...
