Substrate profiling of human Transglutaminase 1 using cDNA display and next-generation sequencing
Biosci Biotechnol Biochem. 2024 Mar 13:zbae029. doi: 10.1093/bbb/zbae029. Online ahead of print.ABSTRACTHuman transglutaminase 1 (TG1) modulates skin development while its involvement in diseases remains poorly understood, necessitating comprehensive exploration of its substrate interactions. To study the substrate profile of TG1, an in vitro selection system based on cDNA display technology was used to screen two peptide libraries with mutations at varying distance from the reactive glutamine. Next-generation sequencing and bioinformatics analysis of the selected DNA pools revealed a detailed TG1 substrate profile indicat...

Convenient pre-separation method depends on negative charges suitable for glycan sequencing analysis
Biosci Biotechnol Biochem. 2024 Mar 8:zbae030. doi: 10.1093/bbb/zbae030. Online ahead of print.ABSTRACTFor precise structural analysis of complex glycans, it is necessary to fractionate glycans based on acidity prior to liquid chromatography-mass spectrometry (LC-MS) analysis. Conventional fractionation using high performance liquid chromatography (HPLC) is time consuming and not high throughput. Therefore, we developed a rapid method for the fractionation using a small cartridge column. This enables rapid multi-sample processing for glycan sequencing.PMID:38458347 | DOI:10.1093/bbb/zbae030 (Source: Bioscience, Biotechnolo...

Regulation of sexual differentiation initiation in Schizosaccharomyces pombe
Biosci Biotechnol Biochem. 2024 Mar 6:zbae019. doi: 10.1093/bbb/zbae019. Online ahead of print.ABSTRACTThe fission yeast Schizosaccharomyces pombe is an excellent model organism to explore cellular events owing to rich tools in genetics, molecular biology, cellular biology, and biochemistry. S. pombe proliferates continuously when nutrients are abundant, but arrests in G1 phase upon depletion of nutrients such as nitrogen and glucose. When cells of opposite mating types are present, cells conjugate, fuse, undergo meiosis, and finally form four spores. This sexual differentiation process in S. pombe has been studied extensi...

Human milk-specific fat components enhance the secretion of ghrelin by MGN3-1 cells
In this study, we showed that 1-oleoyl-2-palmitoylglycerol (OP) increased ghrelin secretion, whereas 1,3-dioleoyl-2-palmitoylglycerol (OPO), a major component of human milk TAGs, did not increase ghrelin levels in the ghrelin-secreting cell line, MGN3-1. Therefore, diacylglycerol OP may directly contribute to the regulation of ghrelin secretion. We also found that 2-palmitoylglycerol and 1- and 2-oleoylglycerol increased ghrelin secretion. Finally, we demonstrated that the intracellular cAMP levels and preproghrelin and ghrelin O-acyl transferase expression levels were enhanced by OP treatment in MGN3-1 cells. This may rep...

Cell-free translation system with artificial lipid-monolayer particles as a unique tool for characterizing lipid-monolayer binding proteins
Biosci Biotechnol Biochem. 2024 Mar 5:zbae026. doi: 10.1093/bbb/zbae026. Online ahead of print.ABSTRACTMethods for functional analysis of proteins specifically localizing to lipid monolayers such as rubber particles and lipid droplets are limited. We have succeeded in establishing a system in which artificially prepared lipid monolayer particles are added to a cell-free translation system to confirm the properties of proteins that specifically bind to lipid monolayers in a translation-coupled manner.PMID:38444196 | DOI:10.1093/bbb/zbae026 (Source: Bioscience, Biotechnology, and Biochemistry)
