Anti-cancer effects of plant-derived Micromonospora sp. M2 against A549 and MCF-7 cell lines
Biosci Biotechnol Biochem. 2024 Apr 4:zbae036. doi: 10.1093/bbb/zbae036. Online ahead of print.ABSTRACTThe huge diversity of secondary bioactive metabolites, such as antibiotic and anticancer compounds produced by Micromonospora sp., makes it an attractive target for study. Here, we explored the anti-proliferative activities of Micromonospora sp. M2 extract (MBE) in relation to its pro-oxidative activities in A549 and MCF7 cell lines. Anti-proliferative effects were assessed by treating cells with MBE. We found that treatment with MBE decreased cell proliferation and increased intracellular reactive oxygen species, and tha...

Solubilization and stabilization of lipoic acid trisulfide by creation of various β-cyclodextrin clathrates
Biosci Biotechnol Biochem. 2024 Apr 3:zbae042. doi: 10.1093/bbb/zbae042. Online ahead of print.ABSTRACTLipoic acid trisulfide, a sulfane sulfur-containing trisulfide of α-lipoic acid, holds promise in pharmaceuticals, yet knowledge gaps persist regarding its synthesis, properties, and stability. Here, we synthesized the lipoic acid trisulfide with a purity exceeding 99% from α-lipoic acid on a gram scale and obtained novel β-cyclodextrin clathrates (84-95% yield). Differential scanning calorimetry confirmed the inclusion of lipoic acid trisulfide in β-cyclodextrins. The resulting β-cyclodextrin clathrates exhibited si...

Catechol-O-methyltransferase and monoamine oxidase B inhibitory activities of Australian bee pollen
Biosci Biotechnol Biochem. 2024 Apr 1:zbae041. doi: 10.1093/bbb/zbae041. Online ahead of print.ABSTRACTBee pollen is an apicultural product collected by honeybees from flower stamens and used as a functional food worldwide. In the present study, we aimed to elucidate the functions of Australian bee pollen. Australian bee pollen extracts and their main components were tested for catechol-O-methyltransferase (COMT) and monoamine oxidase B (MAOB) inhibitory activities. These enzymes are key neurotransmitters involved in Parkinson's disease and depression. Myricetin (5), tricetin (6), and luteolin (7) exhibited high COMT inhib...

Chemical modification of Arthrobacter sarcosine oxidase by N-methylisothiazolinone reduces reactivity toward oxygen
In this study, SoxA was chemically modified using MIT (mo-SoxA), and its structural and chemical properties were characterised. Spectral analysis data, oxygen consumption rates, and reactions were compared between intact and mo-SoxA. These demonstrate that the oxidative half-reaction toward oxygen is inhibited by MIT modification. The oxidase activity of mo-SoxA was approximately 2.1% of that of intact SoxA, and its dehydrogenase activity was approximately 4.2 times higher. The C-to-S mutants revealed that cooperative modification of two specific cysteine residues caused a drastic change in the enzyme reaction mode. Based ...

Structure-activity relationship of anti-inflammatory meroterpenoids isolated from Dictyopteris polypodioides in RAW264 cells
In this study, we explored anti-inflammatory compounds from the brown alga Dictyopteris polypodioides and isolated 7 meroterpenoids. Their anti-inflammatory activities were evaluated using the lipopolysaccharide (LPS)-stimulated mouse macrophage cell line, RAW264. Yahazunol (1) exhibited similar nitric oxide (NO) production inhibitory activity as zonarol (2), which has previously been shown to be an anti-inflammatory compound. Yahazunol (1), zonarol (2), and isozonarol (3) inhibited not only nitric oxide production but also inducible nitric oxide synthase (iNOS), interleukin-6 (IL6), and C-C motif chemokine ligand 2 (CCL2)...

Reclassification of a reveromycin-producer and the proposals of Actinacidiphila reveromycinica sp. nov. and Actinacidiphila acidipaludis comb. nov
Biosci Biotechnol Biochem. 2024 Mar 28:zbae032. doi: 10.1093/bbb/zbae032. Online ahead of print.ABSTRACTStreptomyces sp. SN-593, a reveromycin-producer, was previously thought to belong to the genus Streptomyces based on the morphological and chemotaxonomic characteristics. In this paper, we re-considered its taxonomic position according to the current criteria. Phylogenetic analysis using 16S rRNA gene sequences showed that the strain belongs to the genus Actinacidiphila. In multilocus sequence and phylogenomic analyses, the strain SN-593 represented a distinct evolutionary lineage within this genus and its closest neighb...
