Advanced Hollow Cathode Discharge Plasma Treatment of Unique Bilayered Fibrous Nerve Guidance Conduits for Enhanced/Oriented Neurite Outgrowth
Biomacromolecules. 2024 Feb 27. doi: 10.1021/acs.biomac.3c00976. Online ahead of print.ABSTRACTDespite all recent progresses in nerve tissue engineering, critical-sized nerve defects are still extremely challenging to repair. Therefore, this study targets the bridging of critical nerve defects and promoting an oriented neuronal outgrowth by engineering innovative nerve guidance conduits (NGCs) synergistically possessing exclusive topographical, chemical, and mechanical cues. To do so, a mechanically adequate mixture of polycaprolactone (PCL) and polylactic-co-glycolic acid (PLGA) was first carefully selected as base materi...

Structural Consequences of Introducing Bioactive Domains to Designer β-Sheet Peptide Self-Assemblies
Biomacromolecules. 2024 Feb 26. doi: 10.1021/acs.biomac.3c00962. Online ahead of print.ABSTRACTWe applied solid- and solution-state nuclear magnetic resonance spectroscopy to examine the structure of multidomain peptides composed of self-assembling β-sheet domains linked to bioactive domains. Bioactive domains can be selected to stimulate specific biological responses (e.g., via receptor binding), while the β-sheets provide the desirable nanoscale properties. Although previous work has established the efficacy of multidomain peptides, molecular-level characterization is lacking. The bioactive domains are intended to rema...

Enzymatic Synthesis of α-Glucan Microparticles Using Amylosucrases from < em > Bifidobacterium < /em > Species and Its Physicochemical Properties
This study proposes a bottom-up approach for producing GMPs using four amylosucrases from Bifidobacterium sp. (BASs). The physicochemical characteristics of these GMPs were analyzed, and the results showed that the properties of the GMPs varied depending on the type of enzymes used in their synthesis. As common properties, all GMPs exhibited typical B-type crystal patterns and poor colloidal dispersion stability. Interestingly, differences in the physicochemical properties of GMPs were generated depending on the synthesis rate of linear α-glucan by the enzymes and the degree of polymerization (DP) distribution. Consequent...

Water and Oxygen Barrier Properties of All-Cellulose Nanocomposites
Biomacromolecules. 2024 Feb 23. doi: 10.1021/acs.biomac.3c01337. Online ahead of print.ABSTRACTHydroxypropyl cellulose (HPC) is potentially interesting as a biobased, rigid food packaging material, but its stiffness and strength are somewhat low, and its water and oxygen transport rates are too high. To improve these characteristics, we investigated nanocomposites of HPC and cellulose nanocrystals (CNCs). These high-aspect-ratio nanoparticles display high stiffness and strength, and their high crystallinity renders them virtually impermeable. Exchanging the counterions of sulfate-ester decorated CNCs with cetyltrimethylamm...

Aptamer-Targeted Dendrimersomes Assembled from Azido-Modified Janus Dendrimers "Clicked" to DNA
Biomacromolecules. 2024 Feb 23. doi: 10.1021/acs.biomac.3c01108. Online ahead of print.ABSTRACTAmphiphilic Janus dendrimers (JDs), synthetic alternatives to lipids, have the potential to expand the scope of nanocarrier delivery systems. JDs self-assemble into vesicles called dendrimersomes, encapsulate both hydrophobic cargo and nucleic acids, and demonstrate enhanced stability in comparison to lipid nanoparticles (LNPs). Here, we report the ability to enhance the cellular uptake of Janus dendrimersomes using DNA aptamers. Azido-modified JDs were synthesized and conjugated to alkyne-modified DNAs using copper-catalyzed azi...
