Targeting aryl hydrocarbon receptor to prevent cancer in barrier organs
Biochem Pharmacol. 2024 Mar 20:116156. doi: 10.1016/j.bcp.2024.116156. Online ahead of print.ABSTRACTThe skin, lung, and gut are important barrier organs that control how the body reacts to environmental stressors such as ultraviolet (UV) radiation, air pollutants, dietary components, and microorganisms. The aryl hydrocarbon receptor (AhR) is a ligand-dependent transcription factor that plays an important role in maintaining homeostasis of barrier organs. AhR was initially discovered as a receptor for environmental chemical carcinogens such as polycyclic aromatic hydrocarbons (PAHs). Activation of AhR pathways by PAHs lead...

Human recombinant relaxin-2 (serelaxin) regulates the proteome, lipidome, lipid metabolism and inflammatory profile of rat visceral adipose tissue
In conclusion, our results highlight the regulatory role of serelaxin in the VAT proteome and lipidome, lipolytic function, and inflammatory profile, suggesting the implication of several mechanisms supporting the potential benefit of serelaxin for the prevention of obesity and metabolic disorders.PMID:38518995 | DOI:10.1016/j.bcp.2024.116157 (Source: Biochemical Pharmacology)

Sensitization of cancer cells to paclitaxel-induced apoptosis by canagliflozin
Biochem Pharmacol. 2024 Mar 20;223:116140. doi: 10.1016/j.bcp.2024.116140. Online ahead of print.ABSTRACTCancer cells consume more glucose and usually overexpress glucose transporters which have become potential targets for the development of anticancer drugs. It has been demonstrated that selective SGLT2 inhibitors, such as canagliflozin and dapagliflozin, display anticancer activity. Here we demonstrated that canagliflozin and dapagliflozin synergistically enhanced the growth inhibitory effect of paclitaxel in cancer cells including ovarian cancer and oral squamous cell carcinoma cells. Canagliflozin also inhibited gluco...

Biochem Pharmacol. 2024 Mar 19;223:116153. doi: 10.1016/j.bcp.2024.116153. Online ahead of print.ABSTRACTHepatocellular carcinoma (HCC), the most common form of primary liver cancers, accounts for a significant portion of cancer-related death globally. However, the molecular mechanisms driving the onset and progression of HCC are still not fully understood. Emerging evidence has indicated that non-protein-coding regions of genomes could give rise to transcripts, termed non-coding RNA (ncRNA), forming novel functional driving force for aberrant cellular activity. Over the past decades, overwhelming evidence has denoted invo...

Wnt signaling: Modulating tumor-associated macrophages and related immunotherapeutic insights
Biochem Pharmacol. 2024 Mar 19;223:116154. doi: 10.1016/j.bcp.2024.116154. Online ahead of print.ABSTRACTWnt signaling pathways are highly conserved cascades that mediate multiple biological processes through canonical or noncanonical pathways, from embryonic development to tissue maintenance, but they also contribute to the pathogenesis of numerous cancers. Recent studies have revealed that Wnt signaling pathways critically control the interplay between cancer cells and tumor-associated macrophages (TAMs) in the tumor microenvironment (TME) and potentially impact the efficacy of cancer immunotherapy. In this review, we su...

Empagliflozin reduces the adverse effects of diabetes mellitus on testicular tissue in type 2 diabetic Rats: A stereological and biochemical study
Biochem Pharmacol. 2024 Mar 18:116135. doi: 10.1016/j.bcp.2024.116135. Online ahead of print.ABSTRACTEmpagliflozin as an antioxidant decreases blood glucose and insulin resistance in type 2 diabetes mellitus. Base on the empagliflozin antioxidant properties we decided to investigate the its effects on the testis histological changes through stereological techniques and biochemical evaluations in T2 diabetes mellitus rats. Rats were divided into: control, diabetes mellitus (DM, streptozotocin + nicotinamide) and diabetes mellitus + empagliflozin (DM + EMPA, 10 mg/kg/day) groups. 56 days after inducing diabetes mellitus test...

Dbl family RhoGEFs in cancer: Different roles and targeting strategies
Biochem Pharmacol. 2024 Mar 16:116141. doi: 10.1016/j.bcp.2024.116141. Online ahead of print.ABSTRACTSmall Ras homologous guanosine triphosphatase (Rho GTPase) family proteins are highly associated with tumorigenesis and development. As intrinsic exchange activity regulators of Rho GTPases, Rho guanine nucleotide exchange factors (RhoGEFs) have been demonstrated to be closely involved in tumor development and received increasing attention. They mainly contain two families: the diffuse B-cell lymphoma (Dbl) family and the dedicator of cytokinesis (Dock) family. More and more emphasis has been paid to the Dbl family members ...
