Ultra ‐rapid real‐time polymerase chain reaction assay for genotyping of the NT5E gene in Japanese black beef using a mobile PCR device, PCR 1100
AbstractThe concentration of inosine 5 ′-monophosphate (IMP) in beef is an important factor contributing to beef palatability. A previous study suggested that single nucleotide polymorphisms (SNPs) in theecto-5 ′-nucleotidase (NT5E) gene strongly affect the concentration of IMP under postmortem conditions by regulating NT5E enzymatic activity in beef. Genotyping of theNT5E gene is performed using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) or real-time PCR assay. However, these conventional laboratory assays require large installed instruments. They also involve complicated procedures ...

Intake and apparent digestibility of dry matter, milk production, and composition of cows fed with diets containing oilseed cakes: A meta ‐analysis
The objective was to carry out a meta-analysis and principal component analysis to evaluate the effects of cakes on the intake and apparent digestibility coefficient (ADC) of dry matter (DM) and milk production and composition in lactating dairy cows. The data set used in the meta-analysis came from 51 studies published between 2009 and 2019, which resulted in 119 studies with 18 types of cakes evaluated in 1350 cows. Cows fed with cakes increased dry matter intake (DMI) by 0.366 kg d−1 (P < 0.001) and DMI as a function of the animal's body weight by 0.103% (P < 0.0001) compared with the diet without c...

Dynamic changes of intracellular zinc ion level during maturation, fertilization, activation, and development in mouse oocytes
In this study, we evaluated the amounts and localization of intracellular zinc ions during maturation, fertilization, activation, and embryonic development in mouse oocytes. Our results show that abundant zinc ions are present in both immature and mature oocytes. After in vitro fertilization, the amounts of zinc ions were dramatically decreased at the pronuclear (PN) stage. Ar tificial activation by cycloheximide, SrCl2, and TPEN also reduced the amounts of zinc ions in the PN stage. On the other hand, PN embryos derived from sperm injection still showed high level of zinc ions. However, the amounts of zinc ions rapidly in...

Antimicrobial effects of four chemical additives on fermentation quality, aerobic stability, and in vitro ruminal digestibility of total mixed ration silage prepared with local food by ‐products
The objective of the work is to evaluate the effects of four chemical additives on fermentation quality, aerobic stability, and in vitro ruminal digestibility of total mixed ration (TMR) silage. TMR containing 15% spent mushroom substrate, 25% soybean sauce residue, 45% napiergrass (Pennisetum purpureum (L.) Schum.), and 15% concentrate was ensiled with the following: (1) no additives (control), (2) potassium sorbate (PS, 0.1%), (3) sodium benzoate (SB, 0.1%), (4) sodium diacetate (SDA, 0.5%), and (5) calcium propionate (CAP, 0.5%) based on fresh weight. All silos (10 L) were opened for fermentation quality, in vitro rum...

The expression of nutrient chemosensing gate molecules in the ileum and colon is altered for goats fed on a high ‐grain diet
AbstractLimited knowledge is clarified about alterations in the related expression of nutrient chemosensors in the distal small intestine and hindgut under a high-grain (HG) diet in small ruminants. Herein, this study was performed to investigate the expression changes related to nutrient sensing and transport in the ileal and colonic epithelium of goats in response to feeding an HG diet. Twelve Liuyang black goats (similar age and weight) were randomly assigned into two groups: an HG diet (concentrate: hay = 90:10) and a CON diet (concentrate: hay = 55:45). Immunohistochemistry was applied to detect morphological...

Expression and differential posttranscriptional regulation of the elongation factor 1 alpha 1 gene in endometrial caruncle and intercaruncle of Japanese Black cattle at early and mid ‐gestation stages
AbstractThe elongation factor 1 alpha 1 (EEF1A1), an isoform of EEF1A, is one of the most abundant cytoplasmic proteins and an important component of the translational machinery. We investigated the relative expression, alternative polyadenylation (APA), and changes in poly(A) tail length ofEEF1A1 mRNA in the endometrial caruncle (CAR) and intercaruncle (ICAR) at early and mid-gestation in Japanese Black cattle. The relativeEEF1A1 mRNA expression levels in the CAR were the highest on Gestation day 20 and were significantly decreased at mid-gestation. The expression levels in the ICAR were significantly higher than those in...
