The distribution of purinergic receptors, P2Y and P2X, expression within the intestine
Am J Physiol Gastrointest Liver Physiol. 2023 Nov 21. doi: 10.1152/ajpgi.00108.2023. Online ahead of print.ABSTRACTNucleotides are potent extracellular signaling molecules during homeostasis, infection, and injury due to their ability to activate purinergic receptors. The nucleotide ATP activates P2X receptors (P2RXs), while the nucleotides ADP, ATP, UTP and UDP-glucose selectively activate different P2Y receptors (P2RYs). Several studies have established crucial roles for P2 receptors during intestinal inflammatory and infectious diseases, yet the most extensive characterization of purinergic signaling has focused on immu...

Characterization of neuromuscular transmission and projections of muscle motor neurons in the rat stomach
In this study, we recorded neuromuscular transmission to the circular muscle using intracellular microelectrodes to investigate the spread of influence of intrinsic motor neurons. In addition, microanatomical investigations of neuronal projections and pharmacological analysis were conducted to investigate neuromuscular relationships. We found that inhibitory neurotransmission to the circular muscle is graded with stimulus strength and circumferential distance from the stimulation site. The influence of inhibitory neurons declined between 1 and 11 mm from the stimulation site. In the antrum, corpus and fundus the declines a...

< em > Propionibacterium freudenreichii < /em > CIRM-BIA 129 mitigates colitis through S layer protein B-dependent epithelial strengthening
Am J Physiol Gastrointest Liver Physiol. 2023 Nov 21. doi: 10.1152/ajpgi.00198.2023. Online ahead of print.ABSTRACTThe growing incidence of human diseases involving inflammation and increased gut permeability makes the quest for protective functional foods more crucial than ever. Propionibacterium freudenreichii (Pf) is a beneficial bacterium used in the dairy and probiotic industries. Selected strains exert anti-inflammatory effects, and the present work addresses whether the Pf CIRM-BIA129, consumed daily in a preventive way, could protect mice from acute colitis induced by dextran sodium sulfate (DSS), and more precisel...

Cholestenoic Acid as Endogenous Epigenetic Regulator Decreases Hepatocyte Lipid Accumulation in Vitro and in Vivo
This study shows that CA as an endogenous epigenetic regulator decreases lipid accumulation via epigenetic regulation. The results indicate that CA can be considered a potential therapeutic target for the treatment of metabolic disorders.PMID:37961761 | DOI:10.1152/ajpgi.00184.2023 (Source: American Journal of Physiology. Defining a timeline of colon pathologies after keratin 8 loss: Rapid crypt elongation and diarrhea are followed by epithelial erosion and cell exfoliation
Am J Physiol Gastrointest Liver Physiol. 2023 Nov 14. doi: 10.1152/ajpgi.00140.2023. Online ahead of print.ABSTRACTKeratins are epithelial intermediate filament proteins that play a crucial role in cellular stress protection, with K8 being the most abundant in the colon. The intestinal epithelial-specific K8-deficient mouse model (K8flox/flox;Villin-Cre) exhibits characteristics of inflammatory bowel disease, including diarrhea, crypt erosion, hyperproliferation and decreased barrier function. Nevertheless, the order in which these events occur and whether they are a direct cause of K8 loss or a consequence of one event in...
