Albumin promoter-driven FlpO expression induces efficient genetic recombination in mouse liver
CONCLUSIONS: A highly efficient and liver-specific BAC-Alb-FlpO mouse model was developed. In combination with other Cre lines, different genes can be manipulated sequentially in the same cell, or distinct genetic changes can be induced in different cell types of the same organism.PMID:38469630 | DOI:10.1152/ajpgi.00263.2023 (Source: Am J Physiol Gastroi...)

Panaxynol improves crypt and mucosal architecture, suppresses colitis-enriched microbes, and alters the immune response to mitigate colitis
This study builds on our initial data, which showed that panaxynol - a novel, potent, bioavailable compound found in American ginseng - can suppress disease severity in murine colitis. Here we explore the underlying mechanisms by which panaxynol improves both chronic and acute murine colitis. 14-week-old C57BL/6 female mice were either given 3 rounds of dextran sulfate sodium (DSS) in drinking water to induce chronic colitis or 1 round to induce acute colitis. Vehicle or panaxynol (2.5 mg/kg) was administered via oral gavage 3x/week for the study duration. Consistent with our previous findings, panaxynol significantly (p&l...

Protease-activated receptor 2 drives migration in a colon cancer cell line but not in non-cancerous human epithelial cells
Am J Physiol Gastrointest Liver Physiol. 2024 Mar 5. doi: 10.1152/ajpgi.00284.2023. Online ahead of print.ABSTRACTThe inflamed mucosa contains a complex assortment of proteases which may participate in wound healing or in the development of inflammation-associated colon cancer. We sought to determine the role of protease-activated receptor 2 (PAR2) in epithelial wound healing in both untransformed and transformed colonic epithelial cells. Monolayers of primary epithelial cells derived from organoids cultivated from patient colonic biopsies, and of the T84 colon cancer cell line, were grown to confluence, wounded in the pre...

Hepatic LGR4 aggravates cholestasis-induced liver injury in mice
Am J Physiol Gastrointest Liver Physiol. 2024 Mar 5. doi: 10.1152/ajpgi.00127.2023. Online ahead of print.ABSTRACTCurrent therapy for hepatic injury induced by accumulation of bile acid is limited. Leucine-rich repeat G-protein-coupled receptor 4 (LGR4), also known as GPR48, is critical for cytoprotection and cell proliferation. Here, we reported a novel function for the LGR4 in cholestatic liver injury. In the bile duct ligation (BDL) induced liver injury model, hepatic LGR4 expression was significantly down-regulated. Deficiency of LGR4 in hepatocytes (Lgr4LKO) notably decreased BDL-induced liver injury measured by hepat...

A simple and effective genotyping workflow for rapid detection of CRISPR genome editing
Am J Physiol Gastrointest Liver Physiol. 2024 Feb 27. doi: 10.1152/ajpgi.00013.2024. Online ahead of print.ABSTRACTGenetically engineered mouse models play a pivotal role in the modeling of diseases, exploration of gene functions, and the development of novel therapies. In recent years, CRISPR-Cas9-mediated genome editing technology has revolutionized the process of developing such models by enabling precise genome modifications of the multiple interested genes simultaneously. Following genome editing, an efficient genotyping methodology is crucial for subsequent characterization. However, current genotyping methods are la...
