Mitochondrial-derived microprotein MOTS-c attenuates immobilization-induced skeletal muscle atrophy by suppressing lipid infiltration
Am J Physiol Endocrinol Metab. 2024 Jan 3. doi: 10.1152/ajpendo.00285.2023. Online ahead of print.ABSTRACTMOTS-c, a mitochondrial microprotein, has been described as a novel regulator of glucose and lipid metabolism. In addition to its role as a metabolic regulator, MOTS-c prevents skeletal muscle atrophy in high-fat-fed mice. Here, we examined the preventive effect of MOTS-c on skeletal muscle mass using an immobilization-induced muscle atrophy model and explored its underlying mechanisms. Male C57BL/6J mice (10-week-old) were randomly assigned to one of the three experimental groups: non-immobilization control group (ste...

Empagliflozin Improves Vascular Insulin Sensitivity and Muscle Perfusion in Persons with Type 2 Diabetes
Am J Physiol Endocrinol Metab. 2024 Jan 3. doi: 10.1152/ajpendo.00267.2023. Online ahead of print.ABSTRACTSodium glucose cotransporter 2 inhibitors (SGLT2is) improved MACE, heart failure and renal outcomes in large trials; however, a thorough understanding of the vascular physiologic changes contributing to these responses is lacking. We hypothesized that SGLT2i therapy would diminish vascular insulin resistance and improve hemodynamic function, which could improve clinical outcomes. To test this, we treated 11 persons with type 2 diabetes for 12 weeks with empagliflozin 10 mg/d and measured vascular stiffness, endothelial...

Human liver afferent and efferent nerves revealed by 3D/Airyscan super-resolution imaging
Am J Physiol Endocrinol Metab. 2024 Jan 3. doi: 10.1152/ajpendo.00205.2023. Online ahead of print.ABSTRACTNeural regulation of hepatic metabolism has long been recognized. However, the detailed afferent and efferent innervation of the human liver has not been systematically characterized. This is largely due to the liver's high lipid and pigment contents, causing false negative (light scattering and absorption) and false positive (autofluorescence) results in in-depth fluorescence imaging. Here, to avoid the artifacts in 3D liver neurohistology, we embed the bleached human liver in the high-refractive-index polymer for tis...

Vertical sleeve gastrectomy improves glucose-insulin homeostasis by enhancing beta cell function and survival via FGF15/19
Am J Physiol Endocrinol Metab. 2023 Dec 20. doi: 10.1152/ajpendo.00218.2023. Online ahead of print.ABSTRACTVertical sleeve gastrectomy (VSG) restores glucose homeostasis in obese mice and humans. In addition, the increased fibroblast growth factor (FGF)15/19 circulating level post-surgery has been implicated in this effect. However, the impact of FGF15/19 on pancreatic islets remains unclear. Using a diet-induced obese mice model, we demonstrate that VSG attenuates insulin hypersecretion in isolated pancreatic islets, likely due to morphological alterations in the endocrine pancreas such as reduction in islet, beta cell, a...

Monocyte immunometabolic reprogramming in human pregnancy: contribution of trophoblast cells
Am J Physiol Endocrinol Metab. 2023 Dec 20. doi: 10.1152/ajpendo.00357.2023. Online ahead of print.ABSTRACTImmunometabolism research is uncovering the relationship between metabolic features and immune cell functions in physiological and pathological conditions. Normal pregnancy entails a fine immune and metabolic regulation of the maternal-fetal interaction to assist the energetic demands of the fetus with immune homeostasis maintenance. Here, we determined the immunometabolic status of monocytes of pregnant women compared to non-pregnant controls and its impact on monocyte antiinflammatory functions such as efferocytosis...

Reversible expansion of tissue macrophages in response to macrophage colony-stimulating factor (CSF1) transforms systemic lipid and carbohydrate metabolism
CONCLUSION: These results demonstrate the existence of a dynamic equilibrium between CSF1, the mononuclear phagocyte system and control of liver:body weight ratio, which in turn controls systemic metabolic homeostasis. This novel macrophage regulatory axis has the potential to promote fat mobilisation, without changes in appetence, which may have novel implications for managing metabolic syndrome.PMID:38117267 | DOI:10.1152/ajpendo.00347.2023 (Source: Am J Physiol Endocri...)
