Emerging cerebrospinal fluid biomarkers in autosomal dominant Alzheimer's disease
DiscussionEarly abnormalities in CSF tTau, pTau, SNAP-25, VILIP-1, and YKL-40 suggest that synaptic damage, neuronal injury, and neuroinflammation begin shortly after the commencement of brain amyloid accumulation. (Source: Alzheimer's and Dementia: The Journal of the Alzheimer's Association)

Brain arterial dilatation and the risk of Alzheimer's disease
DiscussionUnderstanding the pathophysiology of the association between AD and brain arterial dilatation may reveal new clues to the vascular contributions to AD. (Source: Alzheimer's and Dementia: The Journal of the Alzheimer's Association)

MIND not Mediterranean diet related to 12-year incidence of cognitive impairment in an Australian longitudinal cohort study
DiscussionPreliminary evidence suggests that protective effects of the MIND diet are geographically generalizable. Additional prospective studies are needed in diverse samples to determine the relative effects of the MIND and the Mediterranean diets against cognitive decline. Alteration in sleep architecture and electroencephalogram as an early sign of Alzheimer's disease preceding the disease pathology and cognitive decline
Publication date: Available online 26 February 2019Source: Alzheimer's & DementiaAuthor(s): Feng Zhang, Rujia Zhong, Song Li, Zhenfa Fu, Renfei Wang, Tianxiao Wang, Zhili Huang, Weidong LeAbstractObjectiveThe present work aims to evaluate the significance of sleep disturbance and electroencephalogram (EEG) alteration in the early stage of Alzheimer's disease (AD).Background and RationaleSleep disturbance is common in patients with AD. It is not known if it can occur at the early stage of AD and if EEG recording may help identify the early sign of the disease.Historical EvolutionSleep disturbance in AD has generally been co...

Incidence of dementia after age 90 in a multiracial cohort
DiscussionPatterns of racial/ethnic disparities in dementia seen in younger older adults continue after the age of 90 years, though smaller in magnitude. (Source: Alzheimer's and Dementia: The Journal of the Alzheimer's Association)

Ethnoracial differences in Alzheimer's disease from the FLorida Autopsied Multi-Ethnic (FLAME) cohort
DiscussionDespite a smaller sample size, Hispanics demonstrated longer disease duration with Alzheimer's disease, but not greater lifespan. Neuropathologic differences across ethnoracial groups supported differences in tau pathology distribution and coexisting hippocampal sclerosis, which may impact biomarker studies. Hospitalization, surgery, and incident dementia
DiscussionWe confirm hospitalization as a risk factor for dementia, with repeated hospitalizations a more important risk factor than surgery. (Source: Alzheimer's and Dementia: The Journal of the Alzheimer's Association)

Staging biomarkers in preclinical autosomal dominant Alzheimer's disease by estimated years to symptom onset
DiscussionDuration of each preclinical stage can be estimated in ADAD, facilitating better planning of prevention trials with the EYO cutoffs under the recently released FDA guidance. The generalization of these results to sporadic AD warrants further investigation. (Source: Alzheimer's and Dementia: The Journal of the Alzheimer's Association)

GeneMatch: A novel recruitment registry using at-home APOE genotyping to enhance referrals to Alzheimer's prevention studies
DiscussionGeneMatch, the first trial-independent research enrollment program designed to recruit and refer cognitively healthy adults to AD prevention studies based in part on APOE test results, provides a novel mechanism to accelerate prescreening and enrollment for AD prevention trials. Clinical and cortical decline in the aphasic variant of Alzheimer's disease
DiscussionPreferential language network neurodegeneration is present in PPA irrespective of amyloid status. Clinical and anatomical progression appears to differ for PPA due to Alzheimer's disease versus non–Alzheimer's disease neuropathology, a distinction that may help to inform prognosis and the design of intervention trials. Genetic origin of a large family with a novel PSEN1 mutation (Ile416Thr)
We report a family with a novel variant in PSEN1.MethodsWe performed clinical and genetic evaluation of 93 related individuals from a Colombian admixed population. 31 individuals had whole-genome sequencing.ResultsGenetic analysis revealed a missense variant in PSEN1 (NM_000021.3: c.1247T>C p.Ile416Thr), which originated on an African haplotype and segregated with AD logarithm of the odds score of 6. Their clinical phenotype is similar to sporadic AD except for earlier age at onset: the mean age at onset for mild cognitive impairment was 47.6 years (standard deviation 5.83) and for dementia 51.6 years (standard deviation...
