Biosafety of mesoporous silica nanoparticles; towards clinical translation
Adv Drug Deliv Rev. 2023 Aug 11:115049. doi: 10.1016/j.addr.2023.115049. Online ahead of print.ABSTRACTMesoporous silica nanoparticles (MSNs) have attracted the attention of chemists, who have developed numerous systems for the encapsulation of a plethora of molecules, allowing the use of mesoporous silica nanoparticles for biomedical applications. MSNs have been extensively studied for their use in nanomedicine, in applications such as drug delivery, diagnosis, and bioimaging, demonstrating significant in vivo efficacy in different preclinical models. Nevertheless, for the transition of MSNs into clinical trials, it is im...

A holistic analysis of the intrinsic and delivery-mediated toxicity of siRNA therapeutics
Adv Drug Deliv Rev. 2023 Aug 9:115052. doi: 10.1016/j.addr.2023.115052. Online ahead of print.ABSTRACTSmall interfering RNAs (siRNAs) are among the most promising therapeutic platforms in many life-threatening diseases. Owing to the significant advances in siRNA design, many challenges in the stability, specificity and delivery of siRNA have been addressed. However, safety concerns and dose-limiting toxicities still stand among the reasons for the failure of clinical trials of potent siRNA therapies, calling for a need of more comprehensive understanding of their potential mechanisms of toxicity. This review delves into th...

Emerging Strategies to Bypass Transplant Rejection via Biomaterial-Assisted Immunoengineering: Insights from Islets and Beyond
Adv Drug Deliv Rev. 2023 Aug 5:115050. doi: 10.1016/j.addr.2023.115050. Online ahead of print.ABSTRACTNovel transplantation techniques are currently under development to preserve the function of impaired tissues or organs. While current technologies can enhance the survival of recipients, they have remained elusive to date due to graft rejection by undesired in vivo immune responses despite systemic prescription of immunosuppressants. The need for life-long immunomodulation and serious adverse effects of current medicines, the development of novel biomaterial-based immunoengineering strategies has attracted much attention ...

Progress on the pathological tissue microenvironment barrier-modulated nanomedicine
Adv Drug Deliv Rev. 2023 Aug 5:115051. doi: 10.1016/j.addr.2023.115051. Online ahead of print.ABSTRACTImbalance in the tissue microenvironment is the main obstacle to drug delivery and distribution in the human body. Before penetrating the pathological tissue microenvironment to the target site, therapeutic agents are usually accompanied by three consumption steps: the first step is tissue physical barriers for prevention of their penetration, the second step is inactivation of them by biological molecules, and the third step is a cytoprotective mechanism for preventing them from functioning on specific subcellular organel...
