Cytokine and chemokine profiles in women with endometriosis, polycystic ovary syndrome, and unexplained infertility
Cytokine. 2024 Mar 30;178:156588. doi: 10.1016/j.cyto.2024.156588. Online ahead of print.ABSTRACTNumerous factors (including immunological, congenital, hormonal, and morphological disorders) can lead to infertility. In this regard, 3 specific diseases associated with infertility are discussed in this review study (i.e., polycystic ovary syndrome [PCOS], endometriosis [EMS], and unexplained infertility [UI]). PCOS is a common endocrine disorder characterized by chronic low-grade inflammation, and EMS is a benign disease characterized by the presence of ectopic endometrial tissue. UI refers to couples who are unable to conce...

GSE252198 Nanopore-based full-length transcriptome sequencing of yak (Bos grunniens) testis
Contributors : Xingdong Wang ; Shaoke Guo ; Jie Pei ; Xian GuoSeries Type : Expression profiling by high throughput sequencingOrganism : Bos grunniens ; Bos grunniens x Bos taurusAlternative splicing (AS) is strictly regulated during cell differentiation and development. AS events are common in the testis, but the splicing regulation in spermatogenesis is still unclear. In this experiment, the third-generation ONT sequencing was used to sequence the full-length transcriptome of testicular tissue, and 40,038 new transcripts were obtained, and the proportion was almost close to the number of known transcripts identified....

GSE226551 SRSF1-mediated alternative splicing is required for spermatogonial stem cell differentiation and mitotic-to-meiotic transition [LACE-seq]
Contributors : Wen-Long Lei ; Zongchang Du ; Ruibao SuSeries Type : OtherOrganism : Mus musculusAlternative splicing (AS) plays significant roles in fundamental biological activities. AS also are prevalent in the testis, but the regulations of alternative splicing in spermatogenesis is vague. Here, we report that Serine/arginine-rich splicing factor 1 (SRSF1), plays critical roles in alternative splicing and male reproduction. Male germ cell-specific deletion of Srsf1 led to complete infertility and abnormal spermatogenesis. We further demonstrated that Srsf1 is required for spermatogonial stem cell differentiation and ...

Where the chromatin undergoes major changes, including meiosis-specific spatiotemporal arrangements and remodelling, the establishment of chromatin loop –axis structure, pairing, and crossing over between homologous chromosomes, any deficiencies in these events may induce ...

GSE262471 Gene regulatory activity associated with PCOS reveals DENND1A-dependent testosterone production
We report the discovery of gene regulatory mechanisms that help explain genetic association with PCOS in the GATA4, FSHB and DENND1A loci. We based those findings using a combination of high throughput reporter assays, CRISPR-based epigenome editing, and genetic association analysis from PCOS case and control populations. In addition, we found that elevated endogenous DENND1A expression causes elevated testosterone levels, a hallmark PCOS phenotype. These results further highlight the potential for combining genetic variant analyses with experimental approaches to fine map genetic associations with disease risk. (Source: G...

Individualized Weight Management and Its Impact on Pregnancy Outcomes in Overweight/Obese Infertile Women: A Retrospective Study
CONCLUSIONS: Individualized weight management enhances pregnancy outcomes in overweight/obese infertile women. Achieving a reduction in body weight by 10% or more, combined with effective control of visceral fat, proves important in improving pregnancy outcomes. Excess visceral fat emerges as an adverse factor impacting successful pregnancy.PMID:38551442 (Source: Alternative Therapies in Health and Medicine)

A Meta-analysis of the Efficacy and Safety of Traditional Chinese Medicine in Treating Endometriosis
CONCLUSION: The meta-analysis results confirmed that TCM treatment exhibited better efficacy in managing endometriosis, resulting in higher pregnancy rates, lower abortion rates, and a reduced probability of adverse reactions. These findings not only hold promise for improving the overall quality of life for patients but also offer practical implications for clinicians. The positive outcomes suggest that integrating TCM into endometriosis management could be considered as part of a comprehensive approach to enhance treatment effectiveness and minimize adverse effects, thereby providing clinicians and patients with valuable...

The significant improvement in ovarian PCOS syndrome using hydralazine and alendronate aromatase inhibitor FDA-approved drugs in Wistar rat models
Biomed Pharmacother. 2024 Mar 28;174:116504. doi: 10.1016/j.biopha.2024.116504. Online ahead of print.ABSTRACTPolycystic ovary syndrome (PCOS) is the most common cause of anovulatory infertility. The aim of this study was to investigate the therapeutic potential of vitamin C, glutamine, mesalazine, hydralazine, and alendronate as new drug candidates for the treatment of letrozole-induced PCOS in female Wistar rats. PCOS was induced in rats by intramuscular injection of estradiol valerate (2 mg/kg body weight for 28 days). The rats then received normal saline (PCOS group), letrozole (0.5 mg/kg), vitamin C (100 mg/kg), gluta...
