Perinatal compromise affects development, form, and function of the hippocampus part one; clinical studies
Pediatr Res. 2024 Mar 22. doi: 10.1038/s41390-024-03105-7. Online ahead of print.ABSTRACTThe hippocampus is a neuron-rich specialised brain structure that plays a central role in the regulation of emotions, learning and memory, cognition, spatial navigation, and motivational processes. In human fetal development, hippocampal neurogenesis is principally complete by mid-gestation, with subsequent maturation comprising dendritogenesis and synaptogenesis in the third trimester of pregnancy and infancy. Dendritogenesis and synaptogenesis underpin connectivity. Hippocampal development is exquisitely sensitive to perturbations du...

Continuity of mental disorders in children with chronic physical illness
We examined the prevalence and predictors of homotypic and heterotypic continuity of mental disorder in children with CPI. A sample of 263 children aged 2 –16 years with physician-diagnosed CPI were recruited from outpatient clinics (e.g., dermatology, respiratory) at a Canadian pediatric academic hospital and followed for 24 months. Parent and child-reported mental disorders (mood, anxiety, behavioral, attention-deficit hyperactivity disorder [ADHD ]) were assessed using the Mini International Neuropsychiatric Interview for Children and Adolescents at baseline, 6, 12, and 24 months. Marginal regression models were compu...

mTOR hyperactivity and < em > RICTOR < /em > amplification as targets for personalized treatments in malignancies
Pathol Oncol Res. 2024 Mar 7;30:1611643. doi: 10.3389/pore.2024.1611643. eCollection 2024.ABSTRACTThe increasing knowledge of molecular alterations in malignancies, including mutations and regulatory failures in the mTOR (mechanistic target of rapamycin) signaling pathway, highlights the importance of mTOR hyperactivity as a validated target in common and rare malignancies. This review summarises recent findings on the characterization and prognostic role of mTOR kinase complexes (mTORC1 and mTORC2) activity regarding differences in their function, structure, regulatory mechanisms, and inhibitor sensitivity. We have recent...

Tinnitus-related increases in single-unit activity in awake rat auditory cortex correlate with tinnitus behavior
Hear Res. 2024 Mar 16;445:108993. doi: 10.1016/j.heares.2024.108993. Online ahead of print.ABSTRACTTinnitus is known to affect 10-15 % of the population, severely impacting 1-2 % of those afflicted. Canonically, tinnitus is generally a consequence of peripheral auditory damage resulting in maladaptive plastic changes in excitatory/inhibitory homeostasis at multiple levels of the central auditory pathway as well as changes in diverse nonauditory structures. Animal studies of primary auditory cortex (A1) generally find tinnitus-related changes in excitability across A1 layers and differences between inhibitory neuronal subty...
