Management of pharmacovigilance during the COVID-19 pandemic crisis by the safety department of an academic sponsor: Lessons learnt and challenges from the EU DisCoVeRy clinical trial
Pharmacol Res Perspect. 2023 Jun;11(3):e01072. doi: 10.1002/prp2.1072.ABSTRACTThe current COVID-19 pandemic was an exceptional health situation, including for drug use. As there was no known effective drug for COVID-19 at the beginning of the pandemic, different drug candidates were proposed. In this article, we present the challenges for an academic Safety Department to manage the global safety of a European trial during the pandemic. The National Institute for Health and Medical Research (Inserm) conducted a European multicenter, open-label, randomized, controlled trial involving three repurposed and one-in development d...

WATER POLICY
U.S. wetland protections curtailed
In a decision that reduces federal protections for wetlands, the U.S. Supreme Court last week narrowed the definition of marshy areas covered by the Clean Water Act. A five-justice majority led by Justice Samuel Alito ruled the law applies only to wetlands that have a “continuous surface connection” to nearby regulated waters, rejecting an approach currently used by federal agencies that wetlands require only a “significant nexus.” Four justices led by Justice Brett Kavanaugh argued the new standard is too narrow, ignores the law’s intent to p...

Electrooxidation degradation of hydroxychloroquine in wastewater using a long-acting Ti-based PbO < sub > 2 < /sub > anode with an arc-sprayed (Ti,Zr)N interlayer
Chemosphere. 2023 May 30:139074. doi: 10.1016/j.chemosphere.2023.139074. Online ahead of print.ABSTRACTTo solve the challenges facing the low-cost and high-efficiency purification of water pollution caused by the production and metabolism of biodegradable hydroxychloroquine (HCQ), electrooxidation treatment with a Ti-based PbO2 anode is the most promising technical solution for engineering applications. However, Ti-based PbO2 anodes have apparent disadvantages, such as poor stability and insufficient electrocatalytic activity. To overcome these shortcomings, a novel Ti/(Ti,Zr)N/PbO2 anode was prepared by introducing an arc...

Bisphenol A-induced autophagy ameliorates human B cell death through Nrf2-mediated regulation of Atg7 and Beclin1 expression by Syk activation
Ecotoxicol Environ Saf. 2023 May 29;260:115061. doi: 10.1016/j.ecoenv.2023.115061. Online ahead of print.ABSTRACTThe widely used plasticizer bisphenol A (BPA) is known as an endocrine-disrupting chemical (EDC). Many studies have shown that BPA contributes to diseases involving immune system alterations, but the underlying mechanisms have yet to be elucidated. We previously reported that BPA at concentration of 100 μM caused human B cell death in accordance with an increase in nuclear factor (erythroid-derived 2)-like 2(Nrf2) expression. Autophagy is a cellular process that degraded and recycles cytoplasmic constituents. H...

Hydroxychloroquine-induced Retinal Toxicity
Long-term use of hydroxychloroquine can cause retinopathy, which may result in severe and progressive visual loss. In the past decade, hydroxychloroquine use has markedly increased and modern retinal imaging techniques have enabled the detection of early, pre-symptomatic disease. As a consequence, the prevalence of retinal toxicity in long-term hydroxychloroquine users is known to be higher than was previously estimated. The pathophysiology of the retinopathy is incompletely characterised, although significant advances have been made in understanding the disease from clinical imaging studies. Hydroxychloroquine retinopathy...

Hydroxychloroquine exposure reduces the risk of cardiovasular disease events in patients with hypertension or diabetes mellitus
CONCLUSIONS: HCQ has protective effect on CVD events, including both AMI and ischaemic stroke in the patients with traditional risk factors. The protective effect of HCQ on CVD events is prominent in older patients.PMID:37246776 | DOI:10.55563/clinexprheumatol/y7psev (Source: Clinical and Experimental Rheumatology)
