Role of Alpha-Fetoprotein in the Pathogenesis of Cancer
J Environ Pathol Toxicol Oncol. 2024;43(2):57-76. doi: 10.1615/JEnvironPatholToxicolOncol.2023049145.ABSTRACTAlpha-fetoprotein (AFP) belongs to the albuminoid protein family and is considered as the fetal analog of serum albumin. This plasma protein is initially synthesized in the fetal liver and yolk sac and shows a maximum peak near the end of the first trimester. Later, concentrations begin to decline prenatally and drop precipitously after birth. This protein has three key ligand-binding pockets for interactions with various biomolecules. It contains multiple phosphorylation and acetylation sites for the regulation of ...

Iron metabolism and ferroptosis in Non-alcoholic fatty liver disease: what is our next step?
Am J Physiol Endocrinol Metab. 2024 Mar 20. doi: 10.1152/ajpendo.00260.2023. Online ahead of print.ABSTRACTNon-alcoholic fatty liver disease (NAFLD) is the most common chronic liver disease with increasing prevalence worldwide. NAFLD could develop from simple hepatic steatosis to non-alcoholic steatohepatitis (NASH) causing fibrosis, cirrhosis and even hepatocellular carcinoma. However, the mechanism of NAFLD development was still not be fully defined. Recently, there are emerging evidence showing the dysregulated iron metabolism with elevated serum ferritin and ferroptosis are involved in the NAFLD. The iron metabolism an...

Platelets Affect the Activity of Amino Acid Transporter SNAT4 in HuH-7 Human Hepatoma Cells
In this study, we examined the effects of platelets on the activity of amino acid transporter system A, which is known to be changed by various factors, and we clarified the mechanism by which platelets affect system A activity. Among system A subtypes, we found that sodium-coupled neutral amino acid transporter (SNAT) 4 played a central role in the transport activity of system A in HuH-7 human hepatoma cells. Interestingly, platelets showed a biphasic effect on system A activity: activated platelet supernatants (APS) including the granule contents released from platelets downregulated system A activity at lower concentrat...

Periodic magnetic modulation enhanced electrochemical analysis for highly sensitive determination of genomic DNA methylation
Anal Biochem. 2024 Mar 18:115509. doi: 10.1016/j.ab.2024.115509. Online ahead of print.ABSTRACTDNA methylation aberrations have a strong correlation with cancer in early detection, diagnosis, and prognosis, which make them possible candidate biomarkers. Electrochemical biosensors offer rapid protocols for detecting DNA methylation status with minimal pretreatment of samples. However, the inevitable presence of background current in the time domain, including electrochemical noise and variations, limits the detection performance of these biosensors, especially for low concentration analytes. Here, we propose an ultrasensiti...

NUF2 regulated the progression of hepatocellular carcinoma through modulating the PI3K/AKT pathway via stabilizing ERBB3
In this study, we confirmed a significant increase in NUF2 expression in HCC tissues and established a correlation between high NUF2 expression and adverse outcomes in HCC patients. Through in vitro and in vivo experiments, we demonstrated that genetic inhibition of NUF2 suppressed the proliferation of HCC cells and disrupted the cell cycle. Further investigation into the molecular mechanisms revealed that NUF2 interacted with ERBB3, inhibiting its ubiquitination degradation, thus activating the PI3K/AKT signaling pathway and influencing cell cycle regulation. Overall, this study revealed the crucial role of NUF2 in promot...
