Lack of mTORC2 signalling in CD11c+ myeloid cells inhibits their migration and ameliorates experimental colitis
J Leukoc Biol. 2024 Apr 23:qiae084. doi: 10.1093/jleuko/qiae084. Online ahead of print.ABSTRACTThe Mammalian Target of Rapamycin (mTOR) pathway plays a key role in determining immune cells function through modulation of their metabolic status. By specific deletion of Rictor in CD11c+ myeloid cells (referred to here as CD11cRicΔ/Δ), this study investigated the role of mTOR complex 2 (mTORC2) signalling in dendritic cells (DCs) function in mice. We showed that upon DSS-induced colitis, lack of mTORC2 signalling CD11c+ cells diminishes colitis score, and abrogates dendritic cell (DC) migration to the mesenteric lymph nodes ...

Comprehensive Analysis of Immune Signatures in Primary Biliary Cholangitis and Autoimmune Hepatitis
In conclusion, our analyses reveal that PBC and AIH share similarities and differences regarding to immune profiles. And γδ T, TIGIT+Vδ2 T, and Tfh1 cell frequencies are potential noninvasive immunological markers that can differentiate PBC from AIH.PMID:38652703 | DOI:10.1093/jleuko/qiae085 (Source: Journal of Leukocyte Biology)

Absence of evidence of transfusion transmission risk of Creutzfeldt-Jakob disease in the United States: Results froma 28-year lookback study
We report on a 28-year lookback study of recipients of blood from donors who subsequently developed CJD.METHODS: Patients with diagnosed CJD and a history of blood donation were identified. Blood centers were asked to provide information about the distribution of the donations and consignees were requested to provide information about the recipients of the donations. Vital status of each available recipient was determined and, if deceased, the reported cause(s) of death were obtained primarily from the National Death Index. All recipients included in the study database contributed person-time up to the last recorded review...

Multi-phased kinetics and interaction of protein kinase signaling in glycoprotein VI-induced platelet αIIbβ3 integrin activation and degranulation
CONCLUSION: Kinetic analysis of GPVI-initiated multisite protein phosphorylation in human platelets demonstrates multiple phases and interactions of tyrosine and serine/threonine kinases with activation-altering feedforward and feedback loops partly involving PKC. The protein kinase inhibitor effects on multisite protein phosphorylation and functional readouts reveal that the signaling network of Syk, Btk and PKC controls platelet granule exocytosis and αIIbβ3 integrin activation.PMID:38653482 | DOI:10.1055/a-2311-0117 (Source: Thrombosis and Haemostasis)

A machine learning approach to predict mortality due to immune-mediated thrombotic thrombocytopenic purpura
CONCLUSION: The USTMA Mortality Index was acceptable for predicting mortality due to acute iTTP in the USTMA registry, but not sensitive enough to rule out death. Identifying patients at high risk of iTTP-related mortality may help individualize care and ultimately improve iTTP survival outcomes. Further studies are needed to provide external validation. Our model is one of many recent examples where machine learning models may show promise in clinical prediction tools in healthcare.PMID:38651093 | PMC:PMC11033197 | DOI:10.1016/j.rpth.2024.102388 (Source: Thrombosis and Haemostasis)

A Retrospective Analysis of BCR-ABL-1 Kinase Domain Mutations in Frontline TKI Resistant Chronic Myeloid Leukemia Patients: A Single Centre Experience
AbstractCML is a commonly diagnosed  myeloproliferative neoplasm in India. Tyrosine kinase inhibitors (TKIs) are the current standard of care for management of CML. Mutations in Tyrosine kinase Domain (TKD) result in resistance to TKIs. The aim of this study is to evaluate the pattern of TKD mutations in CML patients having inadeq uate response or resistance to first line TKIs and to analyse the outcome of CML patients with and without mutations. It is a retrospective observational study. Medical records of 1633 CML patients from year 2014 to 2021 were analysed. Out of these 108 patients (6.6%) lost their response or d...

Emerging therapeutic frontiers in cancer: insights into posttranslational modifications of PD-1/PD-L1 and regulatory pathways
AbstractThe interaction between programmed cell death ligand 1 (PD-L1), which is expressed on the surface of tumor cells, and programmed cell death 1 (PD-1), which is expressed on T cells, impedes the effective activation of tumor antigen-specific T cells, resulting in the evasion of tumor cells from immune-mediated killing. Blocking the PD-1/PD-L1 signaling pathway has been shown to be effective in preventing tumor immune evasion. PD-1/PD-L1 blocking antibodies have garnered significant attention in recent years within the field of tumor treatments, given the aforementioned mechanism. Furthermore, clinical research has su...
