IIG Seminar - Why so many ways to Die?
Intracellular lipopolysaccharide (LPS) from Gram-negative bacteria activates caspase-11 causing inflammatory cell death (pyroptosis), IL-1b processing, and lethal septic shock.  How caspase-11 drove these downstream signaling events was largely unknown.  In 2015, we showed that Gasdermin-D (Gsdmd) is essential for caspase-11-dependent pyroptosis and IL-1b release.  Macrophages from Gsdmd – / – mice generated by gene targeting exhibited defective pyroptosis and IL-1b release in response to cytoplasmic LPS or Gram-negative bacteria. Mechanistically, caspase-11 cleaved Gsdmd and the newly generated N-t...

IIG Seminar - Evolution of pathogenic autoantibodies and B cell clones
Autoimmune disease presents a major clinical challenge due to limited treatment options and difficulties predicting adverse events like nephritis and lymphoma. Serum autoantibodies are unreliable prognosticators because they are often detected in patients with mild and severe disease alike. My research combines analysis of clinical samples and mouse models to understand how pathogenic autoantibodies develop and identify therapeutic targets specific to autoreactive B cells. We have developed a multi-omics single cell approach to longitudinally track autoreactive B cell clones harbouring lymphoma mutations in patients with S...

WALS- George Khoury Lecture: Coronavirus Activation and Antagonism of Interferon Signaling Pathways: from MHV to SARS-CoV-2
Coronavirus interactions with the interferon signaling response are crucial for antiviral defense especially at the early stages of infection. Murine coronavirus, MHV, has taught us a lot about the basic biology of this family of viruses as well as antagonism of innate immune response, a hallmark of coronavirus infection. Building on our MHV data, we have more recently worked with human coronaviruses. We have optimized methods for comparing human coronaviruses in a primary nasal cell air liquid interface (ALI) culture system, which models the initial site of respiratory virus infection. Common cold coronaviruses as well as...
