The role of early life origin B cells in adult immunity
The adult immune system is a mosaic of short-lived cells that are continuously replenished by ongoing hematopoiesis and self-sustaining cells that originated from times long gone. Using lineage tracing mediated by CD79aCreERT2 induced at various time points during ontogeny we have identified a neonatal wave of B cell output that is remarkably well preserved into adulthood. In addition to B-1 cells, early-life time-stamped B cells contributed substantially to IgA plasma cells. These were actively maintained by B cell memory within gut chronic germinal centers and contained commensal microbiota reactivity. Interestingly, neo...

Immune evasion of stem cells in health and cancer
The Agudo lab is focused on the study of immune evasion of stem cells in both autoimmunity and cancer. Our goal is to identify targets to protect tissues for regeneration as well as mechanisms to ensure effective responses to immunotherapy in cancer patients. Dr. Agudo has recently discovered a population of quiescent cancer stem cells in breast cancer that survive during T cell attack and lead to resistance to immunotherapy.Air date: 11/29/2023 4:00:00 PM (Source: Videocast - All Events)

NIH ProtIG Seminar by Dr. Lisa Jenkins: Investigation of the Non-Antiviral Effects of an HIV Zinc Finger Inhibitor: Application of Mass Spectrometry to Off-Target Effects
Over the last several years, a class of small molecule inhibitors of the HIV nucleocapsid protein NCp7 have been developed based upon an S-acyl-2-mercaptobenzamide thioester (SAMT) scaffold. These inhibitors covalently modify the zinc-binding domains of NCp7 in vitro and in cells. We have used mass spectrometry to investigate the mechanism of viral inactivation of NC and Gag by SAMT, identifying multiple sites of covalent modification that resulted from SAMT reaction. Recently, we have found that combination of SAMT-247 formulated as a microbicide with a vaccine provided a high degree of protection from viral infection in ...

Genetics at the maternal-placental-fetal interface: insights into fetal growth and cardiometabolic health.
The early life period is critical for long term health. Fetal growth abnormalities and cardiometabolic diseases are interconnected and cause high burden of morbidity. Understanding the complex genetic and environmental factors that underlie these relationships is crucial to developing preventive and therapeutic interventions for maximizing health across the life span. The placenta is critical for fetal development and potentially underlies later onset diseases, but it is understudied. Moreover, to date, perinatal genomic studies have failed to capture human ancestral diversity, which impedes biological understanding of dis...

146th Meeting, National Advisory Council on Drug Abuse
The Council meets three times a year, usually in February, May, and September for 1 day. Typically, the first part of the meeting is a two-hour session devoted to reviewing grant applications, and this is closed to the public. The second part is the open session, and issues such as training, research dissemination, programs, and policy are discussed. The NIDA Director presents scientific and administrative topics in the Director's Report. Council subcommittees report on their progress, and staff and other speakers also present various topics. Please see earlier Council agendas for an appreciation of the breadth of issues c...

NIH Director's Seminar Series: Neural mechanisms of human episodic memory formation
Memory is critical to our everyday experience. We rely upon our memories not only to form our own sense of identity, but also to guide and plan our future actions and behaviors. Understanding the neural mechanisms that underlie human memory formation is therefore critical in order to effectively treat memory disorders which are present in some of the most debilitating yet poorly managed neurological diseases. Our research efforts are focused on investigating the neural correlates of human episodic memory formation by leveraging the opportunities to directly record neural activity across multiple spatial scales from the hum...
