The Protective Effects and Mechanism of Doxepin on Radiation-Induced Lung Injury in Rats
Dose Response. 2022 Jun 7;20(2):15593258221107193. doi: 10.1177/15593258221107193. eCollection 2022 Apr-Jun.ABSTRACTRadiation-induced lung injuries (RILI) is one of the serious complications of radiotherapy posed by the damage of alveolar cells and inflammation over-reaction. We aimed to investigate the potential protective effects of doxepin on RILI (20 Gy total dose at 3 Gy/min of X-ray irradiation), as well as its underlying mechanism. For animal experiments, such parameters as Immunohistochemistry and hematoxylin and eosin (H&E) staining, WBC (white blood cell), CRP (C-reactive protein), Western blot, and q-PCR wer...

In-vitro antiviral activity of doxepin hydrochloride against group B coxsackievirus
In this study, we used a cytopathic effect-based viral inhibition assay to screen an FDA-approved drug library and found that doxepin hydrochloride had potential antiviral activity. Doxepin hydrochloride exhibited strong antiviral activity against coxsackievirus B types 1-3 with a 50% inhibitory concentration of 10.12 ± 0.85 μM. Moreover, doxepin hydrochloride did not exert antiviral activity against other enteroviruses, including enterovirus A71 (subtypes BrCr/C4) and coxsackievirus A (subtypes 6/10/16). Furthermore, doxepin hydrochloride inhibited virus replication in the early stage of the infection cycle rather than ...

Hypnotic and Melatonin/Melatonin-Receptor Agonist Treatment in Bipolar Disorder: A Systematic Review and Meta-Analysis
ConclusionsThere is a paucity of studies examining pharmacological interventions for sleep and circadian rhythm disturbance in BD. Few studies assessed sleep-related symptoms, and none quantitatively examined endogenous melatonin patterns or other circadian rhythms. Melatonin may be a promising candidate for the adjunctive treatment of bipolar mania. However, dose-finding studies and studies with larger sample sizes are needed to confirm its efficacy. We recommend parallel monitoring of sleep and circadian rhythms in future trials. Chronobiology-informed trial designs are needed to improve the quality of future studies.Pro...

A prospective study examining the effect of selected topical and systemic drugs on pruritus grading system score and stat 6 expression in patients of prurigo nodularis
Conclusion: A combination of selected oral and topical agents can effectively control the severity of PN within one month, and this was found to correlate with STAT 6 expression. (Source: Indian Journal of Dermatology)

Pharmacological management of psychoactive substance withdrawal syndrome
ConclusionsSubstitution therapy seems to be the foremost choice for the treatment of psychoactive substance withdrawal. However, other medications are available but poorly utilized. Practitioners can use the armory of other available medications to manage psychoactive substance withdrawal. Patients can broaden their medication choice. Voltage-gated ion channels are a promising niche to explore. (Source: Drugs and Therapy Perspectives)

Changes in histaminergic system in neuropsychiatric disorders and the potential treatment consequences
Curr Neuropharmacol. 2021 Sep 9. doi: 10.2174/1570159X19666210909144930. Online ahead of print.ABSTRACTIn contrast to that of other monoamine neurotransmitters, the association of the histaminergic system with neuropsychiatric disorders is not well documented. In the last two decades, several clinical studies involved in the development of drugs targeting the histaminergic system have been reported. These include the H3R-antagonist/inverse agonist, pitolisant, used for the treatment of excessive sleepiness in narcolepsy, and the H1R antagonist, doxepin, used to alleviate symptoms of insomnia. The current review summarizes ...

The effect of smoking on the plasma concentration of tricyclic antidepressants - a systematic review
CONCLUSION: The effect of smoking on the plasma concentration of TCAs is still not entirely clear. There is a possibility that smoking affects the distribution of TCA metabolites, but this is probably not of clinical importance.PMID:34497000 | DOI:10.1017/neu.2021.28 (Source: Acta Neuropsychiatrica)

Tricyclic antidepressants versus ‘active placebo’, placebo or no intervention for adults with major depressive disorder: a protocol for a systematic review with meta-analysis and Trial Sequential Analysis
DiscussionTricyclic antidepressants are recommended by clinical guidelines and frequently used worldwide in the treatment of major depressive disorder. There is a need for a thorough systematic review to provide the necessary background for weighing the benefits against the harms. This review will ultimately inform best practice in the treatment of major depressive disorder.Systematic review registrationPROSPEROCRD42021226161. (Source: Systematic Reviews)
