Emerging drugs in phase II and III clinical development for the treatment of alcohol use disorder
Expert Opin Emerg Drugs. 2024 Apr 12. doi: 10.1080/14728214.2024.2342951. Online ahead of print.ABSTRACTINTRODUCTION: Alcohol Use Disorder (AUD) poses an ongoing significant global health burden. AUD is highly prevalent and affects not only the individuals with AUD, but also their communities and society at large. Even though pharmacotherapy is an integral part of AUD treatment, the few available substances show limited efficacy and limited clinical impact. Thus, there is a need for new innovative pharmacotherapeutic approaches.AREAS COVERED: This paper provides a comprehensive review of drugs approved for the treatment of...

Doxazosin inhibits vasculogenic mimicry in human non ‑small cell lung cancer through inhibition of the VEGF‑A/VE‑cadherin/mTOR/MMP pathway
In conclusion, the present study demonstrated that doxazosin displayed anti-VM activity in an NSCLC cell model through the downregulation of VEGF-A and VE-cadherin levels, and the suppression of signaling pathways related to the receptor tyrosine kinase, EphA2, protein kinases, AKT and mTOR, and proteases, MMP-2 and MMP-9. These results support the add-on anti-VM effect of doxazosin as a potential agent against NSCLC.PMID:38455663 | PMC:PMC10918514 | DOI:10.3892/ol.2024.14303 (Source: Oncology Letters)

The combination of doxazosin and metyrosine as a preoperative treatment for pheochromocytomas and paragangliomas
ConclusionThe combination of metyrosine and doxazosin as a preoperative treatment for PPGLs affects intraoperative circulatory hemodynamics, such as a reduced occurrence of blood pressure elevation during surgery. Further research is necessary to identify patients who will benefit most from this combination treatment. (Source: Endocrine)

Giant Negative T Waves in an Elderly Patient with Generalized Pain and Weakness
A 72-year-old male with a history of hypertension and chronic kidney disease presented to our emergency department complaining of numbness in his arms and legs, diarrhea, generalized pain, and weakness for 10 days, which gradually worsened and made moving difficult. His medications included trichloromethiazide, azosemide, doxazosin, nifedipine, carvedilol, and cilostazol. Additionally, prednisone had been used to treat cholesterol embolisms in the toes. Approximately 1 month prior, osteoporosis due to chronic kidney disease and steroid use was suspected, and denosumab therapy and calcium replacement were initiated. (Source...
