Cost-effectiveness analysis of selexipag for the combined treatment of pulmonary arterial hypertension
Conclusion: In China, adding selexipag may not be cost-effective for patients with PAH who failed to control their condition after combined treatment of ERA and PDE5i. Results of the analysis can aid discussions on the value and position of selexipag for the combined treatment of PAH. (Source: Frontiers in Pharmacology)

A pilot phase Ib study to evaluate tadalafil to overcome immunosuppression during chemoradiotherapy for IDH-wild-type glioblastoma
CONCLUSIONS: Concurrent tadalafil is well tolerated during chemoradiotherapy for GBM. Tadalafil is associated with a reduction of peripheral MDSCs after chemoradiotherapy and increased CD8 T-cell proliferation and activation.PMID:37554225 | PMC:PMC10406429 | DOI:10.1093/noajnl/vdad088 (Source: Cancer Control)

Effect of phosphodiesterase ‐5 inhibition on SystEmic Right VEntricular size and function. A multicentre, double‐blind, randomized, placebo‐controlled trial: SERVE
ConclusionsIn this trial in adults with systemic right ventricles, right ventricular systolic function, exercise capacity and neuro-hormonal activation remained stable over a 3-year follow-up period. No significant treatment effect of tadalafil was observed. Further research is needed to find effective treatment for improvement of ventricular function in adults with systemic right ventricles. E-209 Comparative analysis of the vasodilatory effects of enteral nimodipine and tadalafil in an animal model of cerebral vasospasm
ConclusionCerebral vasospasm could leave neurologic deficit or sequelae even after proper treatment. Therefore, prevention is important. Tadalafil showed preventive effect against cerebral vasospasm and vasodilative effect similar to that of nimodipine. Therefore, tadalafil could be considered an alternative preventive treatment of cerebral vasospasm.Disclosures J. Improvement in Endothelial Function in men taking phosphordiesterase type 5 inhibitors for erectile dysfunction
CONCLUSIONS: Erectile dysfunction and endothelial dysfunction are risk factors for cardiovascular disease. Our study showed that PDE5i improved endothelial function and erectile dysfunction (with a significant correlation). Improving endothelial function could be the mechanism that leads to a reduction in cardiovascular events and death in men with erectile dysfunction treated with PDE5i.PMID:37506991 | DOI:10.1016/j.amjmed.2023.07.010 (Source: Atherosclerosis)

Spectroscopic determination of hydrophobic adulterant tadalafil by aptasensor based ellipsometry
In this study, we propose an aptasensor based on ellipsometry for the detection of PDE5 inhibitors. To enhance the detection specificity for PDE5 inhibitors, we designed an aptamer with a hydrophobic pocket that incorporates a guanine base-rich region and a three-way junction. This design is particularly effective considering the poor aqueous solubility of PDE5 inhibitors. Preliminary results demonstrate that tadalafil detection in various media can be achieved within the range of 1-2000 ng/mL. The limit of detection for the active compound of tadalafil is as low as 1.82 ng/mL.PMID:37454513 | DOI:10.1016/j.talanta.2023.124...
