BME-SIG Lecture with Molly Schoichet
Drug discovery in cancer typically involves screening cells grown on 2-dimensional, hard plastic dishes; however, since human tissue is neither 2D nor plastic, this environment does not sufficiently emulate human disease. To overcome this limitation, we designed materials (i.e., hydrogels) for 3-dimensional cell culture with the goal of screening cells in an environment that mimics that of native tissue. With 3D cell culture, we gain an understanding of both cell invasion and cell viability, thereby providing insights that are inherently limited with traditional 2D cell culture. To achieve a suitable environment, we synthe...

BME-SIG Lecture: Navigating Academia to Biotech CEO
Dr. Laura Niklason is an Anesthesiologist, critical care physician, and a biomedical engineer. Supported by the NIH Medical Scientist Training Program from 1983 – 1988, she obtained her PhD in Biophysics in 1988 and then her MD in 1991. After completing clinical training, she became involved in the nascent field of tissue engineering/regenerative medicine in the mid-1990 ’ s. Niklason was fortunate to gain support for her research from the NIH, as well as several private foundations and the Defense Department, spanning from 1995 through 2022. Niklason ’ s work constantly sought to fuse concepts in physical sciences a...

Engineered biomaterials for lymph node drug delivery and disease modeling enable next-generation approaches in cancer immunotherapy
The advent of the cancer immunotherapy era has created wide-reaching new opportunities for drug delivery and biomaterial technologies to impact cancer therapy. Major barriers to the safer and more effective treatment of cancer using immunotherapy include achieving sufficient drug doses in target tissues at appropriate time scales as well as enabling drug synergies by co-delivery of combination therapies. More effective modeling of immunotherapy effects in disease-relevant preclinical models also represents a critical hurdle to the effective translation of newly developed therapies, that could likewise enable the developmen...

Modeling Vaccine Responses and Tumor Immunity Using Lymph Nodes and Microfluidics
Predicting the response of the immune system to a new vaccine, immunotherapy, or infection remains a grand challenge of biomedical science. Current models of immunity largely rely on in vivo animal studies that are difficult to control and analyze over time, or simple in vitro cultures that lack the spatial organization and cell-cell interactions of the body. Here, I will share our laboratory ’ s development of technology to model immunity outside the body. By combining intact, live cultures of ex vivo lymph node tissue with microfluidic devices, we can test the impact of localized drug delivery or the effect of cocul...

Bioengineering Measurement of Receptor Biomarkers: Towards Cell Signaling Control in Health and Disease
Dysregulated vascularization is a hallmark of many disorders, including cancers, obesity, atherosclerosis, and the list is still growing. However, researchers have yet to achieve the promise of controlling vascularization to improve human health. We believe that Bioengineering, as a discipline, offers two key advantages for achieving this promise: 1) the sensitive tools and methods that have been engineered to quantitatively measure tissue and vascular microenvironments and 2) systems approaches, which integrate measurements and offer predictive insights. These approaches offer novel platforms for drug discovery and clinic...

Sex-specific in vitro models of aortic valve stenosis
Cardiovascular disease is the leading cause of death in both men and women, yet our mechanistic knowledge of the sex-specific molecular and cellular mechanisms that guide cardiovascular disease progression, particularly in women, remain poorly characterized. Studies evaluating disease mechanisms rarely state the sex of cells used for in vitro studies or are performed primarily in male animal models, causing our gap in knowledge. My laboratory uses precision biomaterials as in vitro and in vivo tools to dissect mechanisms that contribute to sexual dimorphisms in cardiovascular diseases, specifically aortic valve stenosis. I...
