Affinity fine-tuning anti-CAIX CAR-T cells mitigate on-target off-tumor side effects
Mol Cancer. 2024 Mar 16;23(1):56. doi: 10.1186/s12943-024-01952-w.ABSTRACTOne of the major hurdles that has hindered the success of chimeric antigen receptor (CAR) T cell therapies against solid tumors is on-target off-tumor (OTOT) toxicity due to sharing of the same epitopes on normal tissues. To elevate the safety profile of CAR-T cells, an affinity/avidity fine-tuned CAR was designed enabling CAR-T cell activation only in the presence of a highly expressed tumor associated antigen (TAA) but not when recognizing the same antigen at a physiological level on healthy cells. Using direct stochastic optical reconstruction mic...

Opisthorchis viverrini excretory-secretory products suppress GLUT8 of cholangiocytes
Parasitol Res. 2024 Mar 16;123(3):161. doi: 10.1007/s00436-024-08184-3.ABSTRACTOpisthorchis viverrini infection and the subsequent bile duct cancer it induces remains a significant public health problem in Southeast Asia. Opisthorchiasis has been reported to cause reduced plasma glucose levels among infected patients. The underlying mechanism for this phenomenon is unclear. In the present study, evidence is presented to support the hypothesis that O. viverrini exploits host cholangiocyte glucose transporters (GLUTs) in a similar manner to that of rodent intestinal nematodes, to feed on unabsorbed glucose in the bile for su...

Novel metabolic biomarker for early detection and diagnosis to the patients with gastric cardia adenocarcinoma
ConclusionsA total of 25 common and stable differential metabolites may hold promise as liquid non-invasive indicators for GCA diagnosis. HER2 may function as a tumor suppressor gene in GCA, as its overexpression is associated with improved survival. The downregulation of bile acid metabolism in GCA may offer valuable theoretical insights and innovative approaches for precision-targeted treatments in GCA patients. (Source: Cancer Medicine)

The role of bile acid metabolism in bone and muscle: from analytics to mechanisms
Crit Rev Clin Lab Sci. 2024 Mar 15:1-19. doi: 10.1080/10408363.2024.2323132. Online ahead of print.ABSTRACTOsteoporosis and sarcopenia are both common age-related disorders that are associated with increased morbidity and mortality. Bone and muscle are metabolically very active tissues that require large amounts of energy. Bile acids (BAs), a group of liver-derived steroid compounds, are primarily known as emulsifiers that facilitate the resorption of dietary fat and lipids. In addition, they have pleiotropic metabolic functions in lipoprotein and glucose metabolism, inflammation, and intestinal bacterial growth. Through t...

Intestinal microbiota and biliary system diseases
DiscussionThe existing research mostly focuses on exploring the mechanisms of bacteria targeting a single disease. Lacking comparison of multiple diseases and changes in bacteria during the disease process. We hope to provide biomarkers forearly diagnosis of biliary system diseases and provide new directions for the mechanism of intestinal microbiota in biliary diseases. Genetically and Clinically Confirmed Atypical Cerebrotendinous Xanthomatosis with Normal Cholestanol and Marked Elevations of Bile Acid Precursors and Bile Alcohols
Cerebrotendinous xanthomatosis (CTX, OMIM 213700) is a rare autosomal recessive disorder associated with deficiency of the CYP27A1 enzyme (sterol 27-hydroxylase), encoded by the CYP27A1 gene1, 2. The presentation of clinical features in CTX is complex, with heterogeneous symptoms, variable severity and evolution of complications, and diverse differences in progression of disease even within the same family1, 2. In a series of 43 cases in the United States, the mean age at diagnosis was 32 years3. In these patients, 53% experienced chronic diarrhea in childhood, 74% had cognitive impairment in childhood adolescence, 70% wer...

Sortilin in biliary epithelial cells promotes ductular reaction and fibrosis during cholestatic injury
This study investigated the interplay between sortilin, IL-6 and LIF in cholestatic injury-induced ductular reaction, morphogenesis of new ducts and fibrosis. Cholestatic injury was induced by bile duct ligation (BDL) in WT and Sort1-/- mice, with or without augmentation of IL-6 or LIF. (Source: American Journal of Pathology)

Safety of skipping the tract dilation step for EUS-guided biliary drainage in patients with unresectable malignant biliary obstruction (with video)
ConclusionEmploying the tract dilation step during EUS-BD was a significant risk factor for early AEs. Metal stents with a delivery diameter  <  7.5 Fr can be inserted directly without the tract dilation step, resulting in lower early AE rates. (Source: Surgical Endoscopy)
