Molecular docking analysis of PPAR γ with phytochemicals from Moroccan medicinal plants
In this study, we extensively investigated Moroccan phytochemicals using computational structure-based screening with the crystal structure of the PPARγ ligand-binding domain (PDB ID: 7awc) to discover novel phytochemicals targeting PPARγ. The docking results of 540 Moroccan phytochemicals were integrated into online databases for further exploitation through in-depth studies. Drug-likeness analysis was performed to assess the phytochemicals drug-like properties. Two promising phytochemicals, 3,4-dicaffeoylquinic acid and Chlorogenic acid, were identified, both exhibiting high docking affinity and unique binding site int...

Epithelial metabolic stress in IPF: Identifying intervention strategies using a preclinical murine SftpcI73T model of lung fibrosis
Disruptions to autophagy, mitochondrial biology, and glycolysis are recognized features of alveolar type 2 (AT2) cell stress associated with human IPF. We aimed to leverage our published inducible murine SftpcI73T (IER-SP-CI73T) model of lung fibrosis to catalogue AT2 metabolic dysfunction, identify therapeutic targets, and gauge effect size from focused rescue of the disease phenotype. Primary SftpcI73T AT2 cells (AT2I73T) exhibit acquired autophagic defects that coincide with transcriptomic upregulation in glucose utilization / carbohydrate metabolism pathways and functional biochemical increases in glucose flux and extr...

Risk of Pancreatitis With Incretin Therapies Versus Thiazolidinediones in the Veterans Health Administration
CONCLUSION AND RELEVANCE: This retrospective cohort study of national VHA data found a relatively low incidence of pancreatitis in both cohorts, and an adjusted odds ratio found no statistical difference of pancreatitis in patients prescribed an incretin therapy compared with a control group. This data adds to growing evidence that incretin therapies do not seem to be associated with an increased risk of developing pancreatitis.PMID:37881914 | DOI:10.1177/10600280231205490 (Source: The Annals of Pharmacotherapy)

Inhibitory effect and mechanism of Rosiglitazone on M1 type polarization of central microglia in intracerebral hemorrhage mice based on JNK/STAT3 signaling pathway
ConclusionRosiglitazone could attenuate the inflammation in ICH through inhibiting microglia M1 polarization. Thus, our research would shed now lights on exploring new therapeutic strategies against ICH. (Source: Brain and Behavior)

At the cellular level, we have shown earlier that rosiglitazone accelerates osteoblast differentiation from human mesenchymal stem cells (hMSC) at the expe...

Head-to-head comparison of BAM15, semaglutide, rosiglitazone, NEN, and calorie restriction on metabolic physiology in female db/db mice
Biochim Biophys Acta Mol Basis Dis. 2023 Oct 2:166908. doi: 10.1016/j.bbadis.2023.166908. Online ahead of print.ABSTRACTMetabolic disorders such as type 2 diabetes, fatty liver disease, hyperlipidemia, and obesity commonly co-occur but clinical treatment options do not effectively target all disorders. Calorie restriction, semaglutide, rosiglitazone, and mitochondrial uncouplers have all demonstrated efficacy against one or more obesity-related metabolic disorders, but it currently remains unclear which therapeutic strategy best targets the combination of hyperglycaemia, liver fat, hypertriglyceridemia, and adiposity. Here...

New role for the anandamide metabolite prostaglandin F < sub > 2 α < /sub > ethanolamide: Rolling preadipocyte proliferation
J Lipid Res. 2023 Sep 18:100444. doi: 10.1016/j.jlr.2023.100444. Online ahead of print.ABSTRACTWhite adipose tissue regulation is key to metabolic health, yet still perplexing. The chief endocannabinoid anandamide metabolite, prostaglandin F2α ethanolamide (PGF2αEA) inhibits adipogenesis, i.e., the formation of mature adipocytes. We observed that adipocyte progenitor cells-preadipocytes-following treatment with PGF2αEA, yielded larger pellet sizes. Thus, we hypothesized that PGF2αEA might augment preadipocyte proliferation. Cell viability MTT and crystal violet assays, cell counting, and 5-bromo-2'-deoxyuridine (BrdU) ...

Rosiglitazone inhibits acyl-CoA synthetase long-chain family number 4 and improves secondary brain injury in a rat model of surgical brain injury
This study aimed to investigate the role of ACSL4 in SBI via the ferroptosis pathway. Ninety male Sprague-Dawley rats were examined using a model of SBI. Subsequently, the inhibitory effect of rosiglitazone on ACSL4 was assessed via western blot, real-time polymerase chain reaction (PCR), immunofluorescence, fluoro-jade C staining, Perl's staining, ROS assay, and neurological scoring. The results showed that compared with the Sham group, the protein levels of ACSL4 and transferrin were significantly increased after SBI. Administration of rosiglitazone significantly reduced neuronal necrosis, iron deposition, brain water co...
