Polygenic Scores for Psychiatric Disorders: One Important Piece of the Risk Prediction Puzzle

In the current issue of Biological Psychiatry, Ni et  al. (1) compare ten polygenic score methods for their ability to predict affection status of individuals diagnosed with either schizophrenia (SCZ) or major depressive disorder (MDD). In particular, their work showcases methodological rigor by evaluating polygenic scores (PGSs) using real data from a set of cohorts from the Psychiatric Genomics Consortium. To evaluate performance, they leave out one cohort to test predictions and use the other cohorts for training and, when required by the method, tuning of parameters.
