Construction of an optically controllable CRISPR-Cas9 system using a DNA origami nanostructure

Chem Commun (Camb). 2021 May 13. doi: 10.1039/d1cc00876e. Online ahead of print.ABSTRACTWe demonstrated the photo-controlled sequence-selective dsDNA cleavage using a DNA origami structure with Cas9 nuclease. The activity of Cas9 incorporated inside the ring-shaped DNA origami was completely suppressed. After photoirradiation, Cas9 was released, and its activity for selective dsDNA cleavage was restored.PMID:33982688 | DOI:10.1039/d1cc00876e
