Although strong evidence indicates that plasmacytoid dendritic cells (pDCs) function in viral immunity, autoimmunity, and immune tolerance, the involvement of this cellular subtype in cross priming T cells in  vivo has been incompletely understood. Using a vaccine model in which antigens are delivered only to pDCs, Fu et al. (2020) found that in vivo pDCs cross- primed naive CD8 T cells to yield antigen-specific CD8 T cells and sustained immunity. Cross priming by the pDCs required the presence of bys tander conventional dendritic cells (cDCs), specifically cDC1s, which were not contacted directly by pDCs.
