ONC201 Induces Apoptosis in Gastric Adenocarcinoma and Exhibits Synergy with rhTRAIL

Gastric adenocarcinoma (GA) has a poor 5-year survival of only 31.5%, partly due to ineffective chemotherapy options. The TNF-related apoptosis-inducing ligand (TRAIL) pathway leads to cell death by binding the death receptor 5 (DR5), inducing apoptosis. ONC201 is a small molecule that activates the TRAIL pathway via the integrated stress response, and has been shown to induce cell death in numerous tumor types. We investigated the ability of ONC201 to induce cell death in GA as monotherapy and in combination with recombinant human TRAIL (rhTRAIL).
