Evaluation of next-generation sequencing and Crystal Digital PCR ™ for chimerism monitoring of post-allogeneic haematopoietic stem cell transplantation.

Haematopoietic stem cell transplantation (HSCT) is a curative treatment for most haematological diseases. To evaluate the level of donor engraftment, chimerism must be carefully monitored after HSCT. Chimerism monitoring is very important in the engraftment phase to detect graft failure but also post-HSCT to detect the risk of disease relapse in malignant disease1,2. Currently, two historical methods, short tandem repeat (STR) PCR and qPCR (quantitative PCR) methods, are routinely performed in laboratories.
