Single-cell lineage tracing approaches in hematology research – technical considerations

Hematopoietic stem and progenitor cells (HSPCs) are responsible for the sustained production of mature blood cells during homeostasis and in response to hematopoietic stresses1,2. Aberrant HSPC activity is also linked to a spectrum of hematological diseases, from clonal hematopoiesis to leukemias3-6. The classical models of hematopoiesis predict the existence of a homogeneous pool of multipotent and self-renewing hematopoietic stem cells (HSCs), which are equally contributing to the balanced production of all blood cell lineages through progressive lineage differentiation7,8.
