We collected single cell from testis from the four groups AOS 0 (control), AOS 10 (AOS 10 mg/kg body weight (BW)), B+A 0 (busulfan alone), B+A 10 (busulfan plus AOS 10 mg/kg BW). The the cells were further captured with 10x Genomics based single-cell system. By using single-cell RNA seq, we found busulfan disrupted spermatogenesis by the dramatical decrease in germ cells. AOS significantly increased the number of germ cells and also promoted the germ cell development. Besides, we provide candidate master regulons regulated by AOS. Our data here offers the opportunity for treating of azoospermia or oligospermia.
