The present study investigated the role of nitric oxide (NO•) and inhibitors of apoptosis (IAPs) during apoptosis induced by Sasa quelpaertensis Nakai extracts (SQE) in p53-wild type (WT) and p53-null HCT116 colon carcinoma cells. Trypan blue exclusion and Annexin V/propidium iodide assays were used to test for antiproliferation, and apoptosis and cell cycle. Griess and reverse transcription-polymerase chain reaction and western blotting assays were carried out to assay NO• production, and to detect the mRNA and protein levels of Bcl-2, PARP and IAPs. A colorimetric assay was utilized to measure the time-dependent increase in caspase-3 activity. A 50-year-old Italian-American male with a past medical history of colon cancer presented to our urology clinic in Fall 2018 with an acute elevation in serum prostate specific antigen (PSA). In 2015, he had been diagnosed with stage III (T4a, pN1, cM0) cancer of the ascending colon and treated with right hemicolectomy followed by seven cycles of adjuvant combination of folinic acid, 5-fluorouracil, and oxaliplatin (FOLFox) therapy. Final pathology was consistent with signet ring cell carcinoma (SRCC) of the ascending colon with regional nodal metastasis.

We report here that colonic inflammation induces IL6 expression primarily in the CD11b+Ly6G+Ly6Clo polymorphonuclear myeloid-derived suppressor cells (PMN-MDSC) in colon. Although both tumor cells, T cells and myeloid cells all express IL6, PMN-MDSCs are the primary cell type that express IL6 in colon carcinoma, suggesting that IL6 up-regulation is a response to inflammation in colon epithelium and tumor microenvironment.
