GSE147571 Classification by a gene expression profile can help to select oxaliplatin- or irinotecan-based regimen for first-line treatment of metastatic colorectal cancer: Translational research of the TRICOLORE study

Contributors : Shin Takahashi ; Kota Ouchi ; Akira Okita ; Keigo KomineSeries Type : Expression profiling by arrayOrganism : Homo sapiensThe purpose of this study is to explore biomarkers related to clinical outcomes of OX and IRI as TR in the TRICOLORE trial, and a mutational analysis of colon-cancer-related genes, immunohistochemical staining, DNA methylation status, and a comprehensive gene expression analysis were performed as translational research (TR)

AbstractPurposeVolume-outcome relationship is well established in elective colorectal surgery for cancer, but little is known for patients managed for obstructive colon cancer (OCC). We aimed to compare the management and outcomes according to the hospital volume in this particular setting.MethodsPatients managed for OCC between 2005 and 2015 in centers of the French National Surgical Association were retrospectively analyzed. Hospital volume was dichotomized between low and high volume on the median number of patients included per center during the study period.ResultsA total of 1957 patients with OCC were managed in 56 c...

This study was designated to explore the role of cancer stem cells (CSCs) during chemically induced mouse colon carcinogenesis (by 1,2- dimethylhydrazine dihydrochloride, DMH) with/or without the treatment with a targeted (anti-COX-2) therapeutic drug, celecoxib. Two experiments were conducted. The first, a short-term, 16-week mouse colon carcinogenesis bioassay, demonstrates the early stages of colon carcinogenesis. The other is a medium-term, 32-week mouse colon cancer experiment that mimics an end point of colon malignancy. Colon tumors were detected in animals after 32 weeks; histopathologically, they varied from benig...

Chemopreventive effects of raw and roasted oat flakes after in vitro fermentation with human faecal microbiota. Int J Food Sci Nutr. 2020 Jun 01;:1-13 Authors: Glei M, Zetzmann S, Lorkowski S, Dawczynski C, Schlörmann W Abstract The aim of the present study was to analyse chemopreventive effects of oat flakes under consideration of processing. Thin and thick flakes were roasted and subjected to an in vitro digestion and fermentation. Fermentation supernatants (FS) were characterised and chemopreventive effects were analysed in LT97 colon adenoma cells. Compared to the fermentation contro...

SummaryWe previously reported that the metastasis-associated in colon cancer-1 (MACC-D1) is overexpressed in colon cancer. However, so far, only a few effective MACC-1 inhibitors have been identified. To discover novel transcriptional inhibitors of MACC-1, we performed a luciferase reporter-based high-throughput screening (HTS). This strategy discovered rottlerin as an inhibitor of MACC-1 transcription was able to reduce cell motility in colon cancer in time- and dose-dependent manners. Wound healing assay indicated that 48-h treatment with 2.5  μM rottlerin impairs wound closure compared to the controls. Electroph...
