An endothelial microRNA-1 –regulated network controls eosinophil trafficking in asthma and chronic rhinosinusitis

Airway eosinophilia is a prominent feature of asthma and chronic rhinosinusitis (CRS), and the endothelium plays a key role in eosinophil trafficking. To date, microRNA-1 (miR-1) is the only microRNA known to be regulated in the lung endothelium in asthma models.

Both the IL-4/IL-13 and TSLP pathways have been proposed to drive type 2 inflammation in preclinical models of allergic disease. However, to date, only blockade of the IL-4/IL-13 pathway has demonstrated clinical efficacy in the treatment of multiple diseases with underlying type 2 signatures, including atopic dermatitis (AD), asthma and chronic sinusitis with nasal polyposis. Here, we perform a thorough mechanistic comparison of the IL-4/IL-13 and TSLP pathways in promoting type 2 inflammation.

We sought to provide a comprehensive analysis of the redundant and distinct roles of IL-4 and IL-13 in type 2 inflammation and report the mechanism of action dupilumab.

Urticaria is characterized by the appearance of hives or angioedema.1 Chronic urticaria is defined by urticarial episodes that last more than 6 weeks, occurring daily or several times per week.2 Although acute urticaria is commonly associated with infections,3 most cases of chronic urticaria have no identifiable cause. Nevertheless, several reports documented chronic urticaria from infectious causes including Helicobacter pylori.4 Studies show association of upper respiratory infections, including sinusitis, with chronic urticaria with remission after antibiotic therapy.
