GSE144221 DNA methylation markers panel can improve prediction of response to neoadjuvant chemotherapy in luminal B breast cancer

Thus, prediction of treatment response is imperative to avoid exposure of potential non-responders to deleterious side effects of NACT. We have performed genome-wide analysis of DNA methylation by XmaI-RRBS and selected CpG dinucleotides differential methylation of which discriminates BC samples with different sensitivity to NACT. With this data, we have developed multiplex methylation sensitive restriction enzyme PCR (MSRE-PCR) protocol for determining the methylation status of genes that distinguish BC samples with different NACT response. Using MSRE-PCR, we analyzed the methylation status of 15 genes in 25 BC biopsy samples obtained prior to treatment, and developed a panel of 10 DNA methylation markers predictive of the response to NACT in patients with luminal B breast cancer. Validation of this panel on 37 pretreatment luminal B breast cancer samples has demonstrated acceptable quality classification (area under the curve,AUC = 0.803). Accuracy of the test system is 73%, with the sensitivity of 60% and specificity of 100%.

ConclusionsThis survey shows a fairly good use of hRT and a moderate practice of PBI in Italy. Some practices like hRT to the chest wall and regional lymph nodes as well as rRT need further verification. Likewise, the management of post-NAC RT is very heterogeneous. Future national clinical collaborative studies are advocated in order to investigate these controversial topics about breast cancer radiotherapy.
