Night Shifts Implicated in Increased Prostate Cancer Risk

Shift work is positively associated with higher levels of prostate-specific antigen (PSA) levels in men between the ages of 40 and 65, according to results of a new study.

Despite recent therapeutic advances, prostate cancer remains a leading cause of cancer-related death. A subset of castration-resistant prostate cancers become androgen receptor (AR) signaling independent and develop neuroendocrine prostate cancer (NEPC) features through lineage plasticity. These NEPC tumors, associated with aggressive disease and poor prognosis, are driven, in part, by aberrant expression of N-Myc, through mechanisms that remain unclear. Integrative analysis of the N-Myc transcriptome, cistrome, and interactome using in vivo, in vitro, and ex vivo models (including patient-derived organoids) identified a l...

Publication date: 15 December 2019Source: Journal of Molecular Structure, Volume 1198Author(s): Bandar A. Babgi, Magda H. Abdellattif, Mostafa A. Hussien, Naser E. EltayebAbstractThe synthesis of the benzoimidazolyl-ferrocene containing compound was achieved by Knovenagel condensation of ferrocene-1-carbaldehyde and (2-benzoimidazolyl) acetonitrile. Single crystal structure of the compound showed intramolecular (hydrogen bonding) NH⋯N interactions, forming an infinite 1D-chain that propagate along the [010] direction. The titled compound exhibits two major energy bands around 360 nm and 515 nm in the UV–vis ran...

This study aims to investigate the antiproliferative activity and potential mechanism of action of the xanthoquinodin JBIR-99, isolated from fungi Parengyodontium album MEXU 30,054 and identified by single-crystal X-ray crystallography. Cytotoxicity of xanthoquinodin was evaluated in a panel of human cancer cells lines and CCD-112-CoN normal colon cells, using the sulforhodamine B assay. PC-3 prostate cancer cells were used in biochemical assays including cell cycle, mitochondrial transmembrane potential (MTP), reactive oxygen species (ROS) and caspase activity. Expression levels of apoptosis-pathway-related proteins were ...

The list of unmet clinical needs in prostate cancer is long. Although the problems of overdiagnosis and overtreatment of prostate cancer are increasingly acknowledged by the medical community, the fact remains that this disease is still the second most common cause of cancer-related deaths in men. Nevertheless, significant progress has been made toward characterizing the complex biology of this disease, and advances in imaging techniques that improve the detection of clinically significant cancers have occurred in recent years.

Conclusions[68Ga]Ga-P16-093 provided diagnostic information that appeared equivalent to [68Ga]Ga-PSMA-11 in this limited series of ten prostate cancer patients presenting with biochemical recurrence, with the kidneys found to be the critical organ. Diminished tracer appearance in the urine represents a potential advantage of [68Ga]Ga-P16-093 over [68Ga]Ga-PSMA-11 for detection of lesions in the pelvis.

In conclusion, our data show how oncogenic and tumor-suppressive drivers of cellular senescence act to regulate surveillance processes that can be circumvented to enable SnCs to elude immune recognition but can be reversed by cell surface-targeted interventions to purge the SnCs that persist in vitro and in patients. Since eliminating SnCs can prevent tumor progression, delay the onset of degenerative diseases, and restore fitness; since NKG2D-Ls are not widely expressed in healthy human tissues and NKG2D-L shedding is an evasion mechanism also employed by tumor cells; and since increasing numbers of B cells express NKG2D ...
