Novel functions of grass carp three p40 isoforms as modulators of Th17 signature cytokine expression in head kidney leukocytes

Publication date: Available online 14 November 2019Source: Fish &Shellfish ImmunologyAuthor(s): Shiyu Feng, Hong Zhou, Yanyan Wang, Xinyang Qiu, Anying Zhang, Xinyan WangAbstractInterleukin (IL)-12p40, a component of IL-12 and IL-23, can be secreted as monomer and homodimer in mammals. Our previous study has proved the existence of natural three p40 isoforms and their proinflammatory properties in grass carp. In the present study, we unexpectedly found that recombinant grass carp p40a/b/c (rgcp40a, rgcp40b and rgcp40c) were able to enhance the mRNA levels of grass carp il-17a/f1 (gcil-17a/f1) in a dose- and time-dependent manner in head kidney leukocytes (HKLs). In agreement with these findings, the enzyme-linked immunosorbent assay (ELISA) showed that rgcp40a, rgcp40b and rgcp40c markedly stimulated gcIl-17a/f1 secretion from the HKLs. Together with their stimulatory effects on grass carp gcil-22 and gcil-26 expression, our data suggested their potential to mediate Th17-like response in grass carp. To support this notion, we investigated the underlying mechanisms for the regulation of rgcp40 isoforms on gcil-17a/f1 expression, and found that three rgcp40 isoforms significantly induced the activation of Erk, Jnk and Stat3 pathways in a time-dependent oscillation in the same cell model. Moreover, three rgcp40 isoforms-induced gcil-17a/f1 mRNA expression was suppressed by the inhibition on Erk, Jnk and Stat3 pathways, suggesting the signaling pathways in the p40 isoforms-me...

RawoolAbstractBrucellosis, caused by Brucella species, is a neglected, under reported and under recognized infection of worldwide distribution. Owing to its greater economic and zoonotic importance, control of brucellosis is a priority target set forth by the WHO and development agencies. The genus Brucella is comprised of 12 species. The four main species infecting humans and livestock populations include B. melitensis, B. abortus, B. suis, and B. canis. Brucellosis is a cause of acute...

When the concentration of n-butanol was 7% (v/v), the Tm value of the melting curve of the 5 strains changed from 89 °...

Conclusion about coagulation nature of growth process of ZrO2 nanoparticles at temperature range where thermal diffusion is still missing was done. The suggestion that widespread models of low-temperature growth of nanoparticles based on collective effects (orientation growth), which is suggest the orienting action of crystalline environment fields as a thermodynamic stimulus of growth processes, are a particular cases of manifestation of the overall coagulation process.

Conclusion: Aprepitant was easily added to the preoperative regimen for pediatric patients who may require it. Our approach limited overuse and subsequent cost concerns. Future studies with a comparator group and a greater sample size are needed to demonstrate its efficacy, especially in comparison to time-honored agents such as ondansetron. No adverse effects were noted in our limited study cohort. PMID: 31819673 [PubMed]

This study estimated healthcare costs and utilisation for patients with AR and asthma. Mean annual outpatient visits, pharmaceutical costs and inpatient hospitalisations were calculated for 2010 and 2014, with pharmaceutical and inpatient costs stratified by AIT use. AR and asthma patients had a 35% higher mean number of physician visits and up to 90% higher mean pharmaceutical costs compared to controls. The cost of pharmaceuticals and inpatient hospitalisations were 54% lower in those prescribed AIT. Further research is recommended to understand the reasons for these cost differences.
