Preparation and in vitro evaluation of Chondroitin sulfate and carbopol based mucoadhesive controlled release polymeric composites of Loxoprofen using factorial design

In current research work, Chondroitin sulfate (Natural), Carbopol-934 (Synthetic) and 2-Acrylamido-2-methylpropane sulfonic based semi-interpenetrating hydrogels were synthesized through free radical polymerization technique using Ethylene Glycol Dimethylacrylate as a cross linker. Nine formulations with different concentrations of Chondroitin sulfate and Carbopol-934 were prepared. Structural and morphological characterization was accomplished by using SEM, FTIR, DSC, TGA and XRD. Chemical evaluation was carried out to explore pH independent swelling characteristics, porosity, drug entrapment efficiency, bioadhesive strength and in-vitro drug release behavior. Results were then statistically analyzed and optimum formulation was selected by using numerical approach of 32 factorial design. Results confirmed the formation of stable and firm hydrogel formulation with pH independent and controlled release of Loxoprofen sodium. These hydrogel formulations can be used as suitable alternative to conventional drug delivery systems to achieve optimum therapeutic outcomes.Graphical abstract
