Staphylococcal Cassette Chromosome mec (SCCmec) Analysis of MRSA. Methods Mol Biol. 2020;2069:59-78 Authors: Yamaguchi T, Ono D, Sato A Abstract Methicillin-susceptible S. aureus changes to methicillin-resistant S. aureus (MRSA) upon the acquisition of staphylococcal cassette chromosome mec (SCCmec), a genomic island that encodes methicillin resistance. SCCmec elements in S. aureus are classified into different types based on the combination of mec gene complexes and ccr gene complexes, which share variations, five classes in mec and eight in ccr. To date, at least 13 types of SCCmec elements have been identified and each SCCmec type has individual characteristics. It is known that hospital-associated MRSA strains carry SCCmec elements of types, I, II, and III, and the majority of community-acquired MRSA strains carry characteristic SCCmec elements, type IV SCCmec or type V SCCmec. New options are urgently needed for the treatment of methicillin-resistant Staphylococcus aureus (MRSA) infections. Balsacone C is a new dihydrochalcone extracted from Populus balsamifera that has been reported previously as being active against Staphylococcus aureus. In this study we demonstrate that the supernatant from novel environmental isolates of Delftia spp. have antimicrobial activity. We characterized the active fraction and identified delftibactin A as a compound with antimicrobial activity. Abstract The in vitro activities of five quinazolinone antibacterials, compounds Q1-Q5, were tested against 210 strains of methicillin-resistant Staphylococcus aureus (MRSA). The MIC50/MIC90 values (both in μg/ml) were as follows: Q1 (0.5/2), Q2 (1/4), Q3 (2/4), Q4 (0.06/0.25) and Q5 (0.125/0.5). Several strains with high MIC values for some of these compounds (from 8 to>32 μg/ml) exhibited amino-acid changes in the penicillin-binding proteins, which are targeted by these antibacterials.
