GSE129525 Introducing a new pre-clinical rat model of advanced NASH by combining carbon tetrachloride with high fat & cholesterol diet

Contributors : R Maeso-Diaz ; J J Lozano ; J Gracia-SanchoSeries Type : Expression profiling by high throughput sequencingOrganism : Rattus norvegicusNASH is a severe form of NAFLD that can progress to cirrhosis and hepatocellular carcinoma and, its incidence has markedly increased in recent years. Current available animal models fail to reflect the whole spectrum of the disease. We herein designed a Barcelona NASH (BarNa) rat model using the combination of high fat&cholesterol diet with CCl4 inhalation for 10 weeks (NASH) or 24 weeks (NASH-CH). This model reflects the full spectrum of human NASH and present the main characteristics of the disease including: steatosis and metabolic syndrome, lipotoxicity, cellular death, inflammation, hepatic fibrosis and portal hypertension. Moreover, the molecular signature of the BarNa model shared the most important pathways involved in the pathophysiology of the human disease.

We hypothesized that the store-operated calcium entry (SOCE) channel, Orai1, participates in the activation of Th17 cells and influences renal injury. In rats, following renal ischemia/reperfusion (I/R), there was a rapid and sustained influx of Orai1+ CD4 T cells and IL-17 expression was restricted to Orai1+ cells. When kidney CD4+ cells of post–acute kidney injury (post-AKI) rats were stimulated with angiotensin II and elevated Na+ (10–7 M/170 mM) in vitro, there was an enhanced response in intracellular Ca2+ and IL-17 expression, which was blocked by SOCE inhibitors 2APB, YM58483/BTP2, or AnCoA4. In vivo, YM...

Ovarian cancer arising from benign endometriotic has been widely reported. cystic adenomyoma is a rare form of adenomyosis. And clear cell carcinoma arising from cystic adenomyosis is even more unusual. Here we report a case of clear cell carcinoma arising from cystic adenomyosis treated with laparoscopic surgery.
