Cancers, Vol. 11, Pages 1198: Clinical-Scale Production of CAR-T Cells for the Treatment of Melanoma Patients by mRNA Transfection of a CSPG4-Specific CAR under Full GMP Compliance Cancers doi: 10.3390/cancers11081198 Authors: Manuel Wiesinger Johannes März Mirko Kummer Gerold Schuler Jan Dörrie Beatrice Schuler-Thurner Niels Schaft Chimeric antigen receptor (CAR)-T cells already showed impressive clinical regressions in leukemia and lymphoma. However, the development of CAR-T cells against solid tumors lags behind. Here we present the clinical-scale production of CAR-T cells for the treatment of melanoma under full GMP compliance. In this approach a CAR, specific for chondroitin sulfate proteoglycan 4 (CSPG4) is intentionally transiently expressed by mRNA electroporation for safety reasons. The clinical-scale protocol was optimized for: (i) expansion of T cells, (ii) electroporation efficiency, (iii) viability, (iv) cryopreservation, and (v) potency. Four consistency runs resulted in CAR-T cells in clinically sufficient numbers, i.e., 2.4 × 109 CAR-expressing T cells, starting from 1.77x108 PBMCs, with an average expansion of 13.6x, an electroporation efficiency of 88.0% CAR-positive cells, a survival of 74.1% after electroporation, and a viability of 84% after cryopreservation. Purity was 98.7% CD3+ cells, with 78.1% CD3+/CD8+ T cells and with minor contaminations of 1.2% NK cells and 0.6% B cells. The resulting CAR-T cells were teste...

ConclusionThe new HSV-1 based platform described provides a potent and versatile approach to developing new oncolytic immunotherapies for clinical use. Each of the modifications employed was demonstrated to aid in optimizing the potential of the virus to both directly kill tumors and to lead to systemic therapeutic benefit. For clinical use, these viruses are expected to be most effective in combination with other anti-cancer agents, in particular PD1/L1-targeted immune checkpoint blockade. The first virus from this program (expressing GALV-GP-R− and hGM-CSF) has entered clinical development alone and in combination ...

The objective is, therefore, to examine the relation between co-morbid CVD at cancer diagnosis and HRQoL among cancer survivors diagnosed with colorectal, thyroid, prostate, endometrium, ovarian cancer, melanoma, (non-)Hodgkin lymphoma, chronic lymphocytic leukemia (CLL), or multiple myeloma (MM) in an exploratory population-based cross-sectional study. Material and methods: Analyses were performed on combined data sets from the PROFILES and Netherlands Cancer Registry (NCR). Data on co-morbid CVD at cancer diagnosis was extracted from the NCR. HRQoL was measured via PROFILES at a median of 4.6 years after cancer di...

Authors: Tattevin P, Tchamgoué S, Belem A, Bénézit F, Pronier C, Revest M Abstract Aseptic meningitis is defined as meningeal inflammation - i.e. cerebrospinal fluid (CSF) pleocytosis≥5 cells/mm3 - not related to an infectious process. Etiologies of aseptic meningitis can be classified in three main groups: (i) systemic diseases with meningeal involvement, which include sarcoidosis, Behçet's disease, Sjögren's syndrome, systemic lupus erythematosus and granulomatosis with polyangiitis; (ii) drug-induced aseptic meningitis, mostly reported with non-steroidal an...

President Donald Trump on Monday signed legislation that will extend the September 11th Victim Compensation Fund until 2092. During the signing ceremony at the White House, Trump was surrounding by first responders, survivors and family members of several who died in the 2001 terrorist attack on the United States. The U.S. House of Representatives and the U.S Senate voted overwhelmingly last week to approve the legislation that will provide a lifetime of assistance for victims and their families. The extension will allow the fund, first established in 2001, to pay for past and future medical claims of those effected by the...

The U.S. Senate followed the House of Representatives on Tuesday and passed legislation that will extend the September 11 Victim Compensation Fund until 2092. The legislation will fund all current and future medical claims of those effected by the 9/11 terrorist attacks on the World Trade Center in New York City, the Pentagon in Washington, and United Flight 93, which crashed in Shanksville, Pennsylvania. The extension of funding will benefit victims who may develop mesothelioma linked to the more than 400 tons of asbestos that covered the 16-acre disaster zone after the twin towers fell. According to World Trade Center R...

Heart cancer: Is there such a thing? Heart cancer is extremely rare. Most cancers found in the heart have come from elsewhere in the body, including breast cancer, kidney cancer, lung cancer, leukemia, lymphoma and melanoma. Cancer and cancer treatments can affect the heart in other ways, as well. AbstractOver 70,000 US young adults are diagnosed with cancer annually, disrupting important life transitions and goal pursuits. Hope is a positive psychology construct associated with better quality of life (QOL) that focuses on goal-oriented thinking. We developed and tested Achieving Wellness After Kancer in Early life (AWAKE), a scalable 8-week app-based program consisting of educational videos, mood/activity tracking, and telephone-based coaching to promote hope and QOL in young adult cancer survivors (YACS, 18 –40 years old). A two-arm RCT was used to examine the feasibility, acceptability, and potential e...
