Combined endoscopic laparoscopic surgical treatment of advanced adenomas and early colon cancer.

CONCLUSIONS: CELS is a feasible treatment for colonic neoplasia where endoscopic resection alone is not technically possible. In case of severe co-morbidity ruling out segmental resection in patients diagnosed with T1 or T2 colorectal cancer, CELS treatment may be considered. FUNDING: none. TRIAL REGISTRATION: This study was assessed by The National Committee on Health Research Ethics (SJ-593), which concluded that the study required no approval from the Committee. The study was approved by the Danish Data Protection Agency (REG-126-2017). . PMID: 31315798 [PubMed - in process]

This study aimed to explore the regulatory role of ELFN1-AS1 in COAD and construct a gene delivery system based on extracellular vesicles (EVs). We found that ELFN1-AS1 levels were obviously increased in COAD patients and COAD tumor cells. Knockdown of ELFN1-AS1 expression by siRNA inhibited COAD cell proliferation and migration. Moreover, silencing ELFN1-AS1 significantly reduced the activation of extracellular signal-regulated protein kinase (Erk), up-regulated the protein expression of E-cadherin and down-regulated vimentin. In addition, we treated human umbilical cord mesenchymal stem cells (hUCMSCs) with siRNA-ELFN1-A...

AbstractIn vivo chemopreventive effects associated with hybrid molecules against colon carcinogenesis remain poorly studied. In a previous study, we showed that styrylcoumarin hybrids3-SC1,7-SC2 (2,7-(4-hydroxy-3,5-dimethoxystyryl)-coumarin), and7-SC3 decrease cell viability of SW480 in a time- and concentration-dependent manner (IC50-SW480/48  h = 6.92; 1.01 and 5.33 µM, respectively) with high selectivity indices after 48 h of treatment (>400; 67.8 and 7.2, respectively). The present study investigates the mechanisms of these three styrylcoumarins to induce cell death, using an...

Authors: Dai D, Zhou B, Zhong Y, Jin H, Wang X Abstract The aim of the present study was to use a competing risk model to analyze the prognostic value of mucinous adenocarcinoma (MAC) in patients with colorectal cancer (CRC). An additional aim was to construct nomograms for estimating the 3- and 5-year overall survival (OS) and cancer specific survival (CSS) rates of patients with primary CRC with MAC. The data were extracted from the Surveillance, Epidemiology, and End Results database, and a Multivariate Cox model and competing risk model were applied to assess the OS and CSS. Cox-based and competing risk-based n...

Conclusions: These studies suggest that in addition to its role as a palliative therapeutic modality, RFA may have clinical potential as an immune-adjuvant therapy by augmenting the efficacy of adoptive T cell therapy. PMID: 31795828 [PubMed - in process]

Authors: Fu C, Zhou N, Zhao Y, Duan J, Xu H, Wang Y Abstract Increasing evidence supports the concept that cancer stem cells (CSCs) are responsible for cancer progression and metastasis, therapy resistance and relapse. In addition to conventional therapies for colon cancer, the development of immunotherapies targeting cancer stem cells appears to be a promising strategy to suppress tumor recurrence and metastasis. In the present study, dendritic cells (DCs) were pulsed with whole-tumor cell lysates or total RNA of CD44+ colon cancer stem cells (CCSCs) isolated from mouse colon adenocarcinoma CT-26 cell cultures and...

Abstract BACKGROUND: Privileged 4H-chromenes possess the potent anticancer and anticonvulsant actives. By inspiring potency of 4H-chromenes and demands of present era of scaffold, discovery of effective molecule was carried out for the treatment of cancer and conversant. OBJECTIVE: Designed and synthesized a novel series of 4H-chromene derivatives from one port synthesis for the treatment of cancer and conversant. METHOD: Substitution of side amide chain was formed in multiple steps on amine group of chromene. Later, anticancer activity of synthesized compounds was investigated against human colon adenoc...

In this study, we got the top 3,075 differentially expressed genes (DEGs) and 1,613 potential prognostic genes by GEPIA 2 and identified 1,166 fitness genes in COAD based on genome-scale CRISPR-Cas9 knockout (GeCKO) screening data. Excluding the genes already reported in the literatures, a total of nine DEGs overlapping with prognostic and fitness genes were further analyzed. High expression of CCT6A, RHOQ, and RRP12 promoted COAD cell growth and were relative to lower survival rate of COAD patients, while high expression of UTP18, DDOST, YRDC, ACTG1, RFT1, and NLE1 also promoted COAD cell growth, but were relative to high...

Conclusions: An algorithm comprising one inpatient or two outpatient ICD-9-CM diagnosis codes for endometrial adenocarcinoma had a high PPV. The results indicate that claims databases can be used to reliably identify cases of endometrial adenocarcinoma in studies seeking a high PPV. PMID: 31781220 [PubMed]

Barrett ’s esophagus (BE) is the only known precursor to esophageal adenocarcinoma (EAC), a cancer associated with a dismal 5-year survival rate (Am J Gastroenterol 2017;112(7):1032-1048). Despite all the advances in this field, we have had minimal impact on the incidence, morbidity and mortality related to this lethal cancer. This has provided the impetus for major gastroenterology societies to propose quality indicators to guide diagnostic, surveillance and treatment strategies including endoscopic eradication therapies and ultimately improve outcomes associated with EAC (Gastroenterology 2015;149 (6):1599-1606, Am...

Conclusions: In patients with two cancers, lung cancer is usually the second event, presumably due to comorbidity, survival and our unique population e.g. radical vs adjuvant or palliative intent. The commonest first cancers were breast and prostate. Prostate, bladder and colon cancer developed lung cancer earlier than breast cancer. These data help to inform which cancer types are likely to develop lung cancer and influence tumour surveillance strategies.
