GSE121319 GPRC5A facilitates cell proliferation and bone metastasis of prostate cancer.

Contributors : Yuuki Imai ; Yuichiro SawadaSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiensProstate cancer is a high frequent disease. Early stage prostate cancer can be cured with high probability by early treatment, but hormone refractory prostate cancer and progressive prostate cancer is poor prognosis. Orphan GPCRs have the potential to become a drug discovery target in various carcinomas, but there are few reports in prostate cancer and there are still many possibilities. By big data analysis, we focused on the biological properties of the prostate cancer cell line and found GPRC5A from Orphan GPCR as one of the regulators of prostate cancer development. GPRC5A knock out by genomic editing in PC3, proliferative ability and bone metastasis was markedly suppressed. In prostate cancer cells, GPRC5A inhibit phosphorylation of CREB and affected the expression of cell cycle related genes. GPRC5A is also involved in the establishment of bone metastases, and correlated with prostate cancer metastasis and prognosis. These dates suggest that GPRC5A facilitates cell proliferation and progression of prostate cancer and can be a target for new prognostic marker or drug discovery of prostate cancer.

In conclusion, our results of bioinformatic analysis, in vitro and in vivo functional analysis suggested that REST may serve as a promoter of metastasis in pancreatic cancer. Introduction Pancreatic cancer, ranked fourth in cancer-related mortality, is characterized by its rapid progression and early metastasis (1). Patients are hardly cured by surgical resection due to the existing local invasion and distant metastasis (2). Elevating the overall survival rate for pancreatic cancer requires further elucidation of the underlying molecular mechanism of its metastasis. Epithelial-mesenchymal transition (EMT), especially...

Kandalaft* Department of Oncology, Center for Experimental Therapeutics, Ludwig Center for Cancer Research, University of Lausanne, Lausanne, Switzerland With the advent of combined immunotherapies, personalized dendritic cell (DC)-based vaccination could integrate the current standard of care for the treatment of a large variety of tumors. Due to their proficiency at antigen presentation, DC are key coordinators of the innate...

Abstract A new polyprenylated polycyclic acylphloroglucinol, garcimultiflorone K (1), has been isolated from the stems of Garcinia multiflora, together with two known compounds, garcimultiflorone A (2) and garcimultiflorone B (3). The structure of new compound 1 was determined through spectroscopic methods including 1D and 2D NMR and MS analyses. The anti-angiogenic and anti-cancer effects of compounds 1-3 were evaluated in human endothelial progenitor cells (EPCs) and cancer cells. Of these, garcimultiflorone K (1) displayed the most potent anti-angiogenic property by suppressing cell growth and tube formation of...

Conclusion: The high specific activities required for the clinical application of these radiopharmaceuticals for prostate cancer diagnosis and therapy require careful control of the ratio activity/precursor as well as incubation temperature and time. 68Ga-PSMA of high RP and SA is being used in patients at our Centre with excellent results. 177Lu-PSMA is ready to start a pilot study with patients.

AbstractNanosomal docetaxel lipid suspension (NDLS) [DoceAqualip] is a novel formulation of docetaxel approved in India for the treatment of breast cancer, hormone-refractory prostate cancer, locally advanced squamous cell carcinoma of the head and neck, non-small cell lung cancer and advanced gastric adenocarcinoma. The lipid-based delivery system of NDLS eliminates the need for polysorbate 80 and ethanol, which are contained in the conventional docetaxel formulation and are associated with hypersensitivity reactions and infusion-related toxicities. Because of the diminished potential for NDLS to cause hypersensitivity re...

Stromally-located androgen receptors (AR) are essential to the biology of the prostate. The role of stromal AR signaling activation in prostate cancer (PCa) is still unclear. Recently, the expression of AR splice variants has been linked to survival of hormone-refractory PCa (HR-PCa). To our knowledge expression of these variants in PCa stroma has not been previously evaluated. Here we have characterized the expression of full length (AR-FL) as well as AR splice variants (AR-Vs) in cancer (CAF) and normal (NPF) prostate fibroblasts and studied their potential role during cancer progression.

Treatments for hormone refractory and metastatic prostate cancer (PC) still remain palliative. Also tumor specific vaccinations when tested in the clinical setting showed results lower than expected. A major limitation to active immunotherapy relies on mechanisms of tolerance adopted by the tumor. Indeed, an immunosuppressive environment is established in PC patients, as well as in the TRAMP mouse model of PC, in which peripheral T cell tolerance to the tumor-associated antigen Tag is acquired early during neoplastic transformation, with mechanisms that still need to be fully clarified.Mast cells (MCs) have been described ...
