A promiscuous kinase inhibitor delineates the conspicuous structural features of protein kinase CK2a1

Protein kinase CK2a1 is a serine/threonine kinase that plays a crucial role in the growth, proliferation and survival of cells and is a well known target for tumour and glomerulonephritis therapies. Here, the crystal structure of the kinase domain of CK2a1 complexed with 5-iodotubercidin (5IOD), an ATP-mimetic inhibitor, was determined at 1.78   Å resolution. The structure shows distinct structural features and, in combination with a comparison of the crystal structures of five off-target kinases complexed with 5IOD, provides valuable information for the development of highly selective inhibitors.

We describe a 24-year-old man with CD who showed an abrupt decline in renal function after administration of UST. Twenty-nine months previously, the patient was diagnosed with CD, and abnormal urinalysis findings in health checkup were coincidentally found at that time. A 64-year old female with a history of a pulmonary embolism on rivaroxaban, breast cancer (mastectomy in 2013), hypertension, and diabetes presented for fatigue for one month. She had intermittent hemoptysis for six months. She was brought to the emergency department complaining of fatigue. Blood pressure was elevated to 165/75 mmHg. On exam, she appeared lethargic with pale conjunctiva and leg edema. She did not have a skin rash. Laboratory studies were significant for creatinine of 15.3 mg/dL, blood urine nitrogen 140 mg/dL, potassium 6.5 mEq/L.
