The antiapoptotic and antioxidative stress effects of Zhisanzhen in the Alzheimer’s disease model rat

Zhisanzhen, a type of acupuncture method, has been commonly used in the treatment of various neurodegenerative disorders in clinics in China. The aim of this study was to confirm the effect of Zhisanzhen on Alzheimer’s disease and the associated mechanism. We used D-galactose and Aβ1-40 injections to establish a rat model of AD. Rats were divided into four groups: normal group, AD group, AD+manual acupuncture (control) group, and AD+manual acupuncture (Zhisanzhen) group. Zhisanzhen was used to treat the AD model rats. We found that Zhisanzhen improved behavioral performance, reduced oxidative stress, increased the neurotransmitter acetylcholine concentration, reduced apoptosis in hippocampal neurons, and down-regulated the expression of apoptosis-related genes and proteins. Conclusions: EA treatment can decrease the expression of hypothalamic CRF and CRF-R1, relieve anxiety and depression, meanwhile reduce the expression of CRF-R1 in the gastrointestinal mucosa, increase ZO-1 expression, and adjust tight junctions (TJs) to repair the intestinal mucosal barrier. The above roles suggest that EA may play a dual role in alleviating the gastrointestinal and psychological symptoms of IBS, suggesting a potentially dual therapeutic role for EA in regulating disorders of gut-brain interaction in IBS rats. In conclusion, this study presents the novel finding that the PVS periductium is composed of squamous MCs. These cells tightly line the luminal surface of the PVS tissue, including PNs, PVs, and small branches of the PVs in the abdominal cavity. These results will help researchers understand the role of the PVS in hyaluronan secretion, tissue repair, inflammation, and tumorigenesis, as well as the fine structure of PVS tissue.
