Construction of the safe neutralizing assay system using pseudotyped Nipah virus and G protein-specific monoclonal antibody.

Construction of the safe neutralizing assay system using pseudotyped Nipah virus and G protein-specific monoclonal antibody. Biochem Biophys Res Commun. 2019 Apr 14;: Authors: Bae SE, Kim SS, Moon ST, Cho YD, Lee H, Lee JY, Shin HY, Lee HJ, Kim YB Abstract Nipah virus (NiV) is a recently emerged paramyxovirus that causes acute respiratory illness and fatal encephalitis in a broad spectrum of vertebrates, including humans. Due to its high pathogenicity and mortality rates, NiV requires handling in biosafety level-4 (BSL-4) containment facilities and no effective vaccines or therapeutic agents are currently available. Since current diagnostic tests for detecting serum neutralizing antibodies against NiV mainly employ live viruses, establishment of more safe and robust alternative diagnostic methods is an essential medical requirement. Here, we have developed a pseudotyped NiV and closely related Hendra virus (HeV) expressing envelope attachment (G) and fusion (F) glycoproteins using the Moloney murine leukemia virus (MuLV) packaging system. We additionally generated polyclonal antibodies (pAbs) against NiV-G and HeV-G and assessed their neutralizing activities for potential utilization in the pseudovirus-based neutralization assay and further application in the serum diagnostic test. To enhance the specificity of neutralizing antibody and sensitivity of the serological diagnostic test, monoclonal antibodies (mAbs) against NiV-G were generated, and among wh...

Irreversible T cell exhaustion limits the efficacy of programmed cell death 1 (PD-1) blockade. We observed that dual CD40-TLR4 stimulation within a single tumor restored PD-1 sensitivity and that this regimen triggered a systemic tumor-specific CD8+ T cell response. This approach effectively treated established tumors in diverse syngeneic cancer models, and the systemic effect was dependent on the injected tumor, indicating that treated tumors were converted into necessary components of this therapy. Strikingly, this approach was associated with the absence of exhausted PD-1hi T cells in treated and distant tumors, while s...

[The Conversation Africa] Despite the availability of vaccines for influenza - or flu - the virus continues to kill people across the world. According to the World Health Organisation, flu epidemics result in 3 million to 5 million cases of severe illness and about 290 000 to 650 000 deaths each year.

CONCLUSION: The preventable causes of Fulminant Acute Hepatitis include hepatitis viruses - primarily the hepatitis A virus - and poisoning. Active vaccination, basic sanitation, and public awareness can reduce the number of patients and, consequently, the costs of liver transplantation due to these causes.RESUMO OBJETIVO: O objetivo desta revis ão foi investigar a epidemiologia da hepatite aguda fulminante na América Latina e Caribe e identificar possíveis ações objetivando melhor compreensão e melhora do suporte desses pacientes. Seley-Radtke Pyrrolo[3,2-d]pyrimidines have been studied for many years as potential lead compounds for the development of antiproliferative agents. Much of the focus has been on modifications to the pyrimidine ring, with enzymatic recognition often modulated by C2 and C4 substituents. In contrast, this work focuses on the N5 of the pyrrole ring by means of a series of novel N5-substituted pyrrolo[3,2-d]pyrimidines. The compounds were screened against the NCI-60 Human Tumor Cell Line panel, and the results were analyzed using the COMPARE algorithm to elucidate potential mechanisms of action. COMPARE analysis returned s...

We report a case of a middle aged seropositive male, virologically well suppressed on second line ART (Anti-Retroviral therapy) who presented with a subacute history of neurological symptoms. On imaging and CSF (cerebrospinal fluid) evaluation, he was found to have CD8 encephalitis - a new, rare but treatable entity. To the best of our knowledge, no case has been reported from India. (Natural News) Earlier in the month, we reported that Antifa fascists had brutally assaulted Quillette journalist Andy Ngo in Portland, Oregon, simply for existing and being conservative. Ngo’s injuries were so severe that he’s actually having to be hospitalized due to severe brain bleed. But as horrific as this incident of leftist domestic terrorism is,...

Since 2016 Cochrane UK has been visiting secondary schools in and around Oxford to talk about Evidence-Based Medicine (EBM) and its relevance in our everyday lives. They started with visiting an after-school Science Club and now boost a growing portfolio of schools who welcome them into their classrooms.  Cochrane UK   have taught pupils from Years 9 -13 in formal lessons and in extracurricular science and medical clubs. Their talks and workshops aim to bring an awareness of why it is important to question the evidence behind medical claims. They use topical newspaper headlines to illustrate how medical evid...
